摘要
以异硫氰酸苯酯(PITC)作为柱前衍生剂,用二元梯度法洗脱,在柱温40℃、紫外检测器254nm条件下,通过柱前衍生-反相高效液相色谱法测定贼小豆中的氨基酸种类及含量。结果表明:贼小豆中17种氨基酸在1.2~250μmol范围内与峰面积比有良好的线性关系,精密度、稳定性良好;加标回收率为97.03%~101.3%,相对标准偏差RSD为0.63%~1.9%(n=3);17种氨基酸干基总含量为194.093-225.714mg/g,贼小豆中氨基酸平均质量分数顺序为谷氨酸〉酪氨酸〉天冬氨酸〉蛋氨酸〉脯氨酸〉精氨酸〉苯丙氨酸〉亮氨酸〉甘氨酸〉赖氨酸〉丙氨酸〉胱氨酸〉组氨酸〉苏氨酸〉缬氨酸〉丝氨酸〉异亮氨酸。该方法简便、准确、高效,测定结果令人满意,为贼小豆食品新资源的食用、药用等开发利用提供参考依据。
To determine the composition and contents of amino acids in Vigna Minima by reversed-phase high performance liquid chromatographic (RP-HPLC) method, an ultimate amino acid column was used, and phenylisothiocyanate (PITC) was the precolumn derivatization agent. Elutions were carried out in a binary gradient mode. The column temperature was 40 %, and detection wavelength was set at 254 nm. The results show that: Totally 17 amino acids were found, the calibration curves of 17 amino acids all exhibited good linearity over the range from 1.2 to 250 μmol. and its precision, stability and repeatability were good, Mean recoveries for 17 amino acids were between 97.03% and 101.3%. with a relative standard deviation (RSD) between 0.63% and 1.9% (n=3), and the total content of amino acids in samples was 194.093-225.714 mg/g. So it has a very good foreground of development of edible. The order of the amino acid contents was Glu〉Tyr〉Asp〉Met〉Pro〉Arg〉Phe〉Leu〉Gly〉Lys〉Ala〉Cys〉His〉Thr〉 Val〉Ser〉lle. This method was simple, accurate and efficient, the measurement results are satisfactory, to provide a theoretical basis for the new resources Vigna Minima edible, medicinal etc.
