摘要
采用体外细胞培养,结合流式细胞术和M T T方法研究汉防己碱对M G63细胞生长的作用特点。汉防己碱(0.33Lg/mL)对M G 63细胞仅有轻度抑制作用,细胞存活率保持在88%—97%。汉防己碱浓度依赖性轻度降低cDDP对MG63细胞的IC50;低浓度T et(0.33Lg/mL)明显提升cDD P对M G63/cDDP细胞I C50,但0.5和1.0Lg/mL显著降低cD DP对M G 63/cDDP细胞的I C50。汉防己碱对M G 63/cDDP细胞则既可增加细胞耐药发展(0.33Lg/mL),也可逆转肿瘤细胞对cDDP的耐药性(1.0Lg/mL)。
(Tet) on the Flow cytometry and MTT method were used to investigate the effects of tetrandrine growth of MG63 and MG63/cDDP cells under the condition of vitro cell culture. It was shown that the cell survivals were maintained at 88%--97% with Tet at the concentration of 0.33μg/mL to 1.0μg/mL. The ICso of cisplatin(cDDP) on MG63 cells decreased with concentration- dependently of Tet; 0. 33μg/mL of Tet increased the ICso of cisplatin(eDDP) on MG63/cDDP cells whereas 0. 5μg/mL and 1.0gg/mL of Tet significantly reduced the IC50 of cisplatin on MG63/cDDP cells. Tet (0. 33 μg/mL) accelerated the resistance of MG63/cDDP cells to cisplatin whereas High concentrations(0. 5 μg/mL and 1.0μg/mL) of Tet reversed that.
