摘要
海藻糖作为一种功能糖,在生命科学、医药、农业、食品工业、化妆品工业等领域都有着广阔的应用前景。长期以来,由于海藻糖制取和提取上存在的困难使海藻糖的价格昂贵。试验通过基因工程技术获得一株具高效表达海藻糖合酶的大肠杆菌,该菌海藻糖合酶产量稳定,产酶酶活高。该大肠杆菌基因工程菌产海藻糖合酶最适温度为50oC,pH7.0-7.5,底物质量浓度为0.3g·mL-1,反应时间为8h,其最高转化率62.14%,每克大肠杆菌干菌体产酶酶活达到311.42U/g。
As a functional sugar, trehalose has a broad application prospects in life sciences, medicine, agriculture, food industry, cosmetics industry and other fields. Because of the difficulties on trehalose production and extraction, the price of trehalose is high. By genetic engineering technology, obtain a recombinant E. coli with high enzyme activity. Through verification testing, the recombinant E. coli has a higher activity, then research its enzymatic conversion conditions for industrial production. The optimum reaction condition of trehalose synthase is pH 7.0-7.5, the substrate con centration of 0.3 g. mL-1, the temperature of 50 ℃, the substrate concentration of 30% and the reaction time of 8 h. The highest conversion rate is 62.14%. Trehalose synthase activity of recombinant E. coli reaches 311.42 U/g.
