摘要
目的:探讨穴位埋线对慢性萎缩性胃炎(CAG)模型大鼠胃黏膜组织Toll样受体(TLR4)和核因子-κB(NF-κB)表达的影响及其可能的作用机制。方法:将60只SD大鼠随机分为4组,正常对照组、模型组、自然恢复组、埋线组,每组15只。除正常对照组外,其余各组均采用自由饮用100 mg/L MNNG配合饥饱失常法复制大鼠CAG模型。造模12周后,正常对照组、模型组处死,自然恢复组只抓取不治疗,埋线组选取中脘、胃俞、足三里进行穴位埋线,每周治疗1次,共治疗8周。治疗结束后观察各组胃黏膜组织病理学及超微结构改变,免疫组化检测TLR4、NF-κB的表达。结果:①与正常对照组比较,各组大鼠胃黏膜组织中TLR4、NF-κB蛋白表达明显增多(P<0.01);②与模型组比较,自然恢复组大鼠胃黏膜组织中TLR4、NF-κB蛋白表达差异无统计学意义(P>0.05);③与模型组比较,埋线组TLR4、NF-κB蛋白表达明显减少(P<0.05或P<0.01);④与自然恢复组比较,埋线组TLR4、NF-κB蛋白表达减少(P<0.05);⑤光镜、电镜观察各组之间也具有显著性区别。结论:穴位埋线可以下调TLR4、NF-κB表达,改善胃黏膜组织损伤,通过抑制炎症反应发挥胃黏膜保护作用。
Objective: To investigate the catgut embedding chronic atrophic gastritis(CAG) model of gastric mucosa Toll-like receptor 4(TLR4) and nuclear factor-kappa B(NF-κB) expression and its possible mechanism.Methods: 60 SD rats were randomly divided into four groups,normal control group,model group,natural recovery group,embedding group,with 15 rats in each group.Except the normal control group,other groups were used the free drinking rats 100 mg / L methyl-nitro-nitrosoguanidine(MNNG) with the hungry eat disorders law CAG model.Normal control group,model group after 12 weeks of successful modeling were sacrificed,natural recovery group after 12 weeks of modeling was with no treatment,embedding select group after 12 weeks of modeling had catgut embedding treatment at Zhongwan,Weiyu,Zusanli,one treatment a week for 8 weeks.Observe mucosal histopathological and ultrastructural changes after the end of treatment,immunohistochemical detection of TLR4 and NF-κB expression.Results: ①Compared with the normal control group,TLR4,NF κB protein expression in gastric mucosa increased significantly(P 0.01);②Compared with model group,in gastric mucosa of TLR4 and NF-κB protein expression natural recovery group were no significant difference(P 0.05);③Compared with model group,TLR4 and NF-κB protein expression in catgut embedding group decreased significantly(P 0.05 or P 0.01);④Compared with natural recovery group,TLR4 and NF-κB protein expression in catgut embedding group decreased(P 0.05);⑤light microscopy and electron microscopy between groups were with a significant difference.Conclusion: Catgut embedding can decrease TLR4 and NF-κB expression,meliorate gastric mucosa injury,and can play a gastric mucosal protective effect by inhibiting the inflammatory response.
