摘要 目的初步探讨胸腺基质淋巴细胞生成素(thymic stromal lymphopoietin,TSLP)在特应性皮炎发病机制中的作用。方法将32只BALB/c小鼠随机分为两组:对照组、实验组,每组16只,以2,4-二硝基氟苯丙酮/橄榄油溶液经皮肤致敏建立小鼠特应性皮炎模型。采用免疫组织化学方法检测小鼠皮损中TSLP的表达。ELISA法测定血清中TSLP,IL-4,IFN-γ及IgE的水平。结果特应性皮炎(atopic dermatitis,AD)小鼠皮损中TSLP阳性表达物质主要位于胞核,胞浆及胞膜无特异性着色,实验组平均光密度值(74.306±17.898)明显高于对照组(46.093±3.347),差异有统计学意义(P<0.05)。AD小鼠血清TSLP,IL-4,IFN-γ及IgE(213.621±34.666,97.831±15.882,70.999±9.604,142.488±16.352)的水平明显高于对照组(151.997±22.328,60.560±9.286,51.224±4.534,14.851±3.971),差异均有统计学意义(P均<0.05)。结论 TSLP在特应性皮炎小鼠血清及皮损中大量表达,在AD的发病中可能发挥重要作用。 Objective To explore the role of thymic stromal lymphopoietin(TSLP) in the pathogenesis of atopic dermatitis. Methods Thirty-two BALB/c mice were randomly divided into control group and experimental group. The atopie dermatitis model was established by topical applications of 2, 4-Dinitrofluorobenzene in acetone/ olive oil. The expression of TSLP in the lesions was evaluated by imnmnohistochemical staining. The serum levels of TSLP, IL-4, IFN-3, and IgE were determined by enzyme-linked immunosorbent assay (ELISA). Results Positive staining was mainly localized in the nucleus of the epidermal cells without specific staining in cytoplasm and membrane. The intensity of TSLP staining in experimental group was signifieanlly higher than that in the controls(74. 306 ± 17. 898 vs. 46. 093 ± 3. 347). The serum levels of TSLP, IL-4, IFN-γ and IgE in experimental group were also higher than that in the controls(213. 621 ± 34. 666 vs. 151. 997 ± 22. 328 for TSLP; 97.831 ± 15. 882 vs. 60. 560 ± 9. 286 for IL-4 ; 70. 999 ± 9. 604 vs. 51. 224 ± 4. 534 for IFN-γ; 142. 488 ±16. 352 vs. 14. 851±3. 971 for IgE). Conclusion The expression levels of TSLP were higher in both AD lesions and serum. TSLP may play a key role in the pathogenesis of atopic dermatitis.
