摘要
目的:制备具有零级释药特征的天麻素缓释片,考察其体外释药影响因素。方法:采用正交试验设计,以片芯药层中促渗剂用量、包衣液中增塑剂用量、包衣增重和释药孔径为考察因素,以12 h内累积释放度和释放度综合评分为指标,筛选最优处方。结果:天麻素缓释片遵从以渗透压作为主要释药动力的释药模式,体外零级释放特征显著,12 h累积释放度达到预期效果。结论:通过处方和工艺优化得到的天麻素缓释片适合产业化生产。
To prepare the gastrodin sustained-release tablets exhibiting zero-order release, and to investigatethe factors influencing the drug release. METHODS By investigating the dosage of penetrating agent in core, the dosage ot plasticiser, the weight of coating membrane and the orifice size, the method of orthogonal design was used to choose the fitting formula with the cumulative percentage of drug release and synthetic scored method as the indexes. RESULTS The preparation complied with the drug release model with the osmotic pressure different as the release kinetic power, which showed the better characteristics of zero order release and had stable release within 12 h. CONCLUSION The optimized preparation is suitable for the industrialized manufacture.
