滇白珠水提物对实验性大鼠慢性阻塞性肺疾病炎症的影响 被引量:4

Ameliorating effects of water extracts fromGaultheria leucocarpa var on inflammation in chronic obstructive pulmonary diseases of rats

摘要 目的:利用滇白珠水提物对慢性阻塞性肺疾病实验性大鼠进行干预,检测其炎症细胞和C-反应蛋白(CRP)、白介素8(IL-8)变化并对其机制进行探讨。方法:健康SD雄性大鼠60只,随机分为5组:正常对照组、模型组和滇白珠水提物低、中、高剂量组(分别为0.5,1.0,2.0g·kg-1),每组12只。选择熏吸香烟加气管内注射脂多糖(LPS)法复制慢阻肺模型,并观察各组大鼠肺组织的病理改变,行支气管肺泡灌洗液细胞计数和白细胞比率测定,测定各组血清和肺组织匀浆中C-反应蛋白、IL-8水平。结果:模型组大鼠出现明显慢性支气管炎特征性病理改变,出现肺气肿,肺泡结构不完整,肺泡壁变薄断裂,气管及血管周围大量炎症细胞浸润,滇白珠水提物低、中、高剂量治疗组也出现肺气肿和炎症浸润,但程度较模型组轻。模型组大鼠肺匀浆组织和血清CRP、IL-8明显较正常对照组增多,高、中、低剂量治疗组大鼠肺匀浆组织和血清CRP、IL-8均明显较模型组减少,差异有显著性(P<0.05或P<0.01);模型组中炎症细胞总数增加,与正常对照组比较差异有极显著性(P<0.01);各剂量治疗组炎症细胞总数较模型组明显减少(P<0.01),差异均有显著性。与正常对照组比较,模型组和治疗组中细胞总数和中性粒细胞比例均显著升高,与模型组比较,高中低剂量治疗组细胞总数和中性粒细胞比例显著下降,并呈一定的剂量效应关系(P<0.01)。结论:滇白珠水提物可降低大鼠肺组织和血清中的C-反应蛋白及IL-8,减轻肺组织及气道炎症,对大鼠的慢阻肺炎症有抑制作用。 OBJECTIVE To investigate the water extracts from Gaultheria leucocarpa var on inflammation in chronic ob structive pulmonary disease of rats, to explore the changes of inflammatory cells and C-reactive protein (CRP), 8 interleukin (IL-8) and the mechanism of actions. METHODS 60 healthy SD male rats were randomly divided into 5 groups as follows: normal control group, model group and Gaultheria leucocarpa var low, middle, high dose treatment groups, 12 rats in each group. The pathological changes of lung were evaluated. The leukocytes count and percentage were detected in bronchoalveolar lavage fluid, the contents of C-reaction protein and IL-8 were measured in serum and lung tissue homogenate. RESULTS The model rats appeared obvious chronic bronchitis pathological characteristic, emphysema, trachea and perivascular infiltration of inflammatory cells; low, medium and high dose treatment group also appeared emphysema and inflammatory infiltration, but the degree was lighter than in model group. In model rats, lung tissue homogenate and serum CRP, IL-8 increased significantly than those in normal control group in the high, middle and low dose treatment rats, lung tissue homogenate and serum CRP, IL-8 were significantly reduced than those in model group, the difference was statistically significant (P〈0. 05 or 0. 01 ). In the model group the total inflammatory cells were increased significantly as compared with normal control group (P〈0. 01). In each dose treatment group the inflammatory cells were obviously reduced than in the model group, with statistically significant difference (P〈0. 01). Comparing with the normal control group, in model group and treatment group the total leukocytes counts and neutrophil percentage were significantly increased, and in the treatment group compared with the model group the total leukocytes counts and neutrophil percentage were decreased significantly, and dose-dependently (P 〈 0.01 ). CONCLUSION Gaultheria leucocarpa var can reduce rats lung tissu
