摘要
番茄腺毛与其防御性反应及次生代谢产物的合成密切相关,为了进一步研究腺毛特异性表达基因,试验比较研究了改良的异硫氰酸胍(GTC)法、CTAB法、Trizol法对番茄腺毛总RNA的提取效果。结果表明,改良GTC法提取的总RNA质量最好,电泳显示条带清晰完整,OD260/OD280比值为1.98。用该法获得的RNA更适合后续分子生物学研究。
Glandular trichomes of tomato are closely related to the plant defense response and synthesis of secondary metabolites. In order to further study on the special expressing gene in plant triehomes, total RNA of glandular trichomes from tomato was extracted with 3 methods, including improved GTC method, CTAB method and Trizol method. The results showed that the high quality RNA was obtained with improved GTC method. Agarose gel electrophoresis showed clear and complete bands, and OD260/OD280 was 1.98. Moreover, it is evident that the total RNA extracted with improved GTC would be suitable for research on molecular biology.
