摘要
目的探讨咖啡酸锗体内抗H22肝癌血管生成的作用及其机制。方法建立荷H22肝癌小鼠动物模型,随机分为5组:生理盐水组(NS组,阴性对照组)、环磷酰胺组(阳性对照组)、咖啡酸锗低、中、高剂量组。尾静脉注射给药,采用体内抗肿瘤法观察咖啡酸锗对小鼠肿瘤组织的影响(抑瘤率),采用HE染色在光镜下观察各组小鼠的肿瘤血管生成情况,以及肿瘤细胞的生长情况,并应用免疫组化SP法检测各组小鼠肿瘤组织中血管内皮生长因子(VEGF)的表达。结果与NS组比较,咖啡酸锗各剂量组瘤重均明显降低(P<0.01),咖啡酸锗低、中、高剂量组的抑瘤率分别为37.78%、48.89%、54.81%,咖啡酸锗各剂量组肿瘤血管数目比NS组明显减少(P<0.01),肿瘤细胞生长受抑,而坏死程度较严重。咖啡酸锗各剂量组肿瘤组织中VEGF的表达明显低于NS组(P<0.05)。结论咖啡酸锗对荷H22肝癌小鼠有明显的抗肿瘤血管生成作用,从而抑制肿瘤的生长,其机制可能与抑制肿瘤组织中VEGF的表达有关。
Aim To explore the antiangiogenic effect of caffeic acid Ge against H22 hepatocellular carcinoma in vivo and its mechanism. Methods Mice bearing H22 hepatocellular carcinoma cells were randomly grouped into 5 groups: normal saline group (NS, neg- ative controlled group ) , cyclophosphamide group (positive controlled group), caffeic acid Ge low, me- dium and high-dose groups, the drug was injected into caudal vein. The tumor inhibitory ratios of caffeic acid Ge were calculated. HE staining and light microscope were employed to observe tumor angiogenesis, tumor proliferation. The expression of vascular endothelial growth factor(VEGF) in the tumor tissue was tested by immunohistochemistry method. Results Compared with NS group, the tumor weight of caffeic acid Ge groups was significantly decreased ( P 〈 0. 01 ) , the tumor inhibitory rates of the low, medium and high- dose groups were 37.78% ,48.89% ,54. 81% respec- tively, the number of blood vessels in the tumor tissue of caffeic acid Ge groups was significantly decreased (P 〈 0.01 ), the tumor proliferation of caffeic acid Ge groups was inhibited, and the necrosis of tumor tissue was worse. VEGF expression in the tumor tissue of caf- feic acid Ge groups was significantly lower than that of NS group ( P 〈 0.05 ). Conclusion Caffeic acid Ge has effective antiangiogenic effects against I-I22 hepato- cellular carcinoma in mice, thereby inhibiting tumor proliferation, its mechanism is probably related to in- hibiting the expression of VEGF in the tumor tissue.
