摘要
目的考察香附挥发油对对乙酰氨基酚体外经大鼠皮肤促渗透作用及香附挥发油经皮透过量,为其作为对乙酰氨基酚的透皮促进剂提供参考。方法建立对乙酰氨基酚和香附挥发油中主要成分α-香附酮含量测定的高效液相色谱(HPLC)法。用改良的Franz扩散池考察α-香附酮和对乙酰氨基酚的体外经皮透过量。结果α-香附酮和对乙酰氨基酚的皮肤渗透行为均符合Higuchi方程,香附挥发油皮肤透过性呈浓度依赖性,1.0%,3.0%,5.0%香附挥发油对对乙酰氨基酚的增渗倍数分别为2.460,3.430,3.078倍,相应浓度氮酮的增渗倍数分别为4.031,11.420,11.180倍。结论香附挥发油具有良好的皮肤渗透性,可以作为对乙酰氨基酚的透皮促进剂。
Objective To study the percutaneous permeability of volatile oil of cyperus rotundus and its enhancement effect on acetaminophen transdermal application in rats in vitro, and provide basis to use as a transdermal enhancer. Methods To establish a method to determine acetaminophen and ot-eyperone by HPLC. In vitro permeation studies were carried out by using modified Franz diffusion cells. Results The transdermal process of α-cyperone and acetaminophen was concordant with Higuehi equation ; The enhancement rates of 1, 0%, 3.0% and 5.0% volatile oil for acetaminophen were 2. 460, 3. 430, and 3,078 times, respectively. The corresponding enhancement rates for Azone were 4. 031, 11. 420, and 11. 180 times, respectively, Conclusion Volatile oil of cyperus rotundus has a good skin permeability, and may be used as transdermal enhancers for acetaminophen.
