摘要
目的 :探讨他莫昔芬对雌二醇抑制大鼠颈总动脉球囊损伤后内膜增生的影响。方法 :8~ 10周龄 SD大鼠 ,分为雄性组和雌性组 ,每组 2 1只 ,各组又平分为 3个亚组 :去势对照组 ,雌二醇组 ,他莫昔芬组。应用 F2 .0 经皮腔内冠状动脉成形术 ( PTCA)球囊导管损伤血管内膜。2周后处死动物 ,取出左、右颈总动脉作切片观察。结果 :雄性组 :内膜面积、内膜面积 /肌层面积值 ,雌二醇组均小于去势对照组 (均 P <0 .0 5 ) ;他莫昔芬组与去势对照组无显著性差异 (均 P >0 .0 5 )。雌性组 :内膜面积、内膜面积 /肌层面积值 ,雌二醇组均小于去势对照组 (均 P <0 .0 5 ) ,而他莫昔芬组与去势对照组相比亦无统计学差异 (均 P >0 .0 5 )。结论 :雌二醇对去势雄性、雌性大鼠颈总动脉损伤后均有抑制内膜增生作用 ;他莫昔芬拮抗雌二醇的作用 。
Objective:To investigate effect of tamoxifen on estradiol inhibition neointimal proliferation after rats carotid common arteries injury. Method:Eight^ten weeks old gonadectomized rats were divided into groups of male (n=21)and female(n=21),in which consists of three subgroups:gonadectomized control (n=7),estradiol (n=7),tamoxifen (n=7). Rats were killed two weeks after their common carotid artery arteries were injured with F 2.0 PTCA balloon catheter.Left and right carotid common arteries were measured. Data were analysed with t test,P< 0.05 considered to be significant.Result: Neointimal areas,ratios of intima/media in estradiol subgroups all were less than those of control subgroups (all P< 0.05 ), those of tamoxifen subgroups all were similar to those of control subgroups(all P> 0.05 ).Conclusion: Estradiol inhibited neointimal proliferation after rats carotid common artery injury.Tamoxifen antagonized effect of estradiol,suggested that estradiol inhibited neointimal proliferation by estrogen receptor.
