摘要
目的 了解基质降解对原发性肝癌的作用。方法 33例原发性肝细胞癌患者术中取癌组织及癌旁组织 ;提取组织总RNA ;用逆转录共扩增定量PCR方法测定明胶酶A (MMP2 )基因表达水平。结果 肝癌组织明胶酶A基因表达水平 (3 2 0± 0 34 )明显高于癌旁的肝硬化组织 (0 70±0 16 )及正常肝组织 (0 34± 0 0 7)。肝癌患者MMP2 基因表达水平与血清AFP浓度、肿瘤大小无关。结论 明胶酶A在原发性肝癌的进展中可能起重要作用。
Objective To investigate the role of extracellular matrix degradation on primary hepatocellular carcinoma. MethodsCancerous and paracancerous tissue were obtained from 33 patients with primary hepatocellular carcinoma undergoing surgery. Total RNA was isolated and gelatinase A (MMP 2) gene expression level were rneasured by reverse transcription polymerase chain reaction (RT PCR).Results[WT5”BZ] The gelatinase A (MMP 2) gene expression level in cancerous tissue(3 20±0 34) was much higher than those of cirrhotic tissue(0 70±0 16) and normal liver tissue(0 34±0 07). MMP 2 gene expression levels has no correlation with serum concentration of AFP and tumor size. Conclusion Gelatinase A may play an important role in the progression of primary hepatocellular carcinoma.
