依托咪酯对大鼠皮质、海马cAMP含量的影响 被引量:1

摘要 目的 研究依托咪酯对大鼠脑cAMP含量及腺苷酸环化酶(AC) 活性的影响。方法 30 只SD 大鼠,随机分为3 组,即对照组、依托咪酯1 组和依托咪酯2 组。对照组等容腹腔注射生理盐水5 ml/kg,依托咪酯1 和2组分别腹腔注射依托咪酯6 mg/kg 和12 mg/kg。前2 组腹腔注射3 min 后断头,后一组翻正反射消失后立即断头。分别采用蛋白竞争结合法及以非标记ATP为底物的方法测SD大鼠各脑区( 皮质、海马、脑干)的cAMP含量与AC活性变化。结果 依托咪酯1 组大鼠行为变迟钝,依托咪酯2 组大鼠翻正反射消失、皮层内cAMP含量升高且与对照组相比以及两剂量组间皆存在统计学差异( P< 0.05)。两组海马cAMP含量均高于对照组( P<0 .05)。依托咪酯对各脑区AC活性变化无明显影响( P > 0.05)。结论 依托咪酯的全身麻醉作用与其升高皮质、海马cAMP的含量有关。 Objective To investigate the effects of etomidate on cAMP level and adenylate cyclase (AC) activity in rat brain. Methods 30 SD rats were divided randomly into 3 groups.The animals were intraperitoneally administered equal volumes of normal saline 5 ml·kg -1 (control group),etomidate 6 mg·kg -1 (etomidate 1 group)、 and etomidate 12 mg·kg -1 (etomidate 2 group), respectively. The rats in etomidate 2 group were decapitated immediately after the disappeareance of righting reflex, while those in control group and etomidate 1 group were decapitated 3 minutes after the injection of normal saline or etomidate.The rapidly removed brain stem, hippocampus and cerebral cortex, were homogenized and centrifuged. The cAMP level was assayed by protein competition binding analysis and the AC activity was analysed by taking non-radiolabelled ATP as the substrate. Results While righting reflex was lost in etomidate 2 group, the rats' be havior looked sluggish in etomidate 1 group. The cortical cAMP level in etomidate 2 group was increased, higher than in control and etomidate 1 group (P<0.05). The level of hippocamp al cAMP was increased in both etomidate groups (P<0.05).However,AC activities in the 2 etomidate groups were not affected (P>0.05). Conclusion The increment of cAMP level in hippocampus and cerebral cortex may be related to the anesthestic effect of etomidate.
