离子交换树脂分离纯化山豆根中苦参碱和氧化苦参碱的实验研究 被引量:5

摘要 目的研究建立离子交换树脂柱纯化分离山豆根中苦参碱和氧化苦参碱方法。方法山豆根经醋酸溶液浸泡水提后,采用732型阳离子交换树脂柱分离纯化。以苦参碱和氧化苦参碱含量为指标,考察以乙醇溶液、盐酸溶液等不同洗脱剂的分离纯化效果。结果经过优选,分离纯化最佳条件为:样品过柱流速为0.7mL/min,水洗至中性后,以2倍柱体积12%盐酸溶液为洗脱剂洗脱杂质,再以100mL 20%盐酸溶液震荡解吸1h。结论优选建立的山豆根离子交换树脂纯化步骤简便实用、成本低,回收效率高,且对环境及操作者无伤害,为山豆根药材有效组分分离和含量检测奠定了基础。 Objective To establish a method for purification the matrine and oxymatrine in Sophorae tonkinesis by ion exchange resin. Methods The purifying process of matrine and oxymatrine from Sophora tonkinensis was performed by ion exchange resin of 732 type and the contents of matrine and oxymatrine were as the index to investigate the separated effects using alcohol solution and hydrochloric acid solution as the eluent. Results The optimal purification conditions were that the flow-rate was 0.7mL/min, and the eluent were water and 12% hydrochloric acid solution. After the residual impurities were eluted, the ion exchange resin that matrine and oxymatrine were adsorbed was oscillation desorpted with 100mL of 20% hydrochloric acid solution for lh. Conclusion The purification process was simple, practical and high efficiency. It was harmless to the operator and environment, and can be used for purifying the matrine and oxymatrine in Sophorae tonkinesis.
