明胶-白芨胶/丹参多孔材料复合大鼠真皮成纤维细胞构建组织工程真皮的实验研究 被引量:1

摘要 目的:探讨明胶-白芨胶/丹参多孔材料复合大鼠真皮成纤维细胞制成组织工程真皮。方法:制备明胶-白芨胶/丹参多孔材料及其浸提液,与L-929细胞共同培养,观察浸提液与L-929细胞相容性。提取大鼠真皮层中成纤维细胞,与材料复合构建组织工程真皮。复合组织采用苏木精-伊红染色及电镜下观察结合效果。结果:L-929细胞在浸提液中生长情况良好。大鼠真皮成纤维细胞与材料共同培养2周,苏木精-伊红染色及电镜观察,细胞能在材料上正常粘附与生长。结论:明胶-白芨胶/丹参多孔材料无细胞毒性。能与大鼠真皮成纤维细胞成功复合为组织工程真皮。 Objective :To compose the tissue-engineering corium by gelatin-bletilla/Salvia-miltiorrhiza material and fibroblast of rat corium. Method:The gelatin-bletilla/ Salvia-miltiorrhiza material and the leaching liquor of material were prepared. The leaching liquor was cultured with L-929 cells. The tissue-engineering corium was constructed by gelatin-bletilla/ Salvi a-miltiorrhiza material combined with fibroblast extracted from rat corium. The effect of combination was observed by TEM and HE staining. Result : Leaching liquor did not cause cytotoxicity to L-929 cells when they were co-cultured. Fibroblast had got normal growth and adhesion in the material after 2 weeks. Conclusion:The gelatin-bletilla/Salvia-miltiorrhiza as porous material has no cytotoxicity. The material can be composed with fibroblast of rat corium as a tissue-engineering corium.
