摘要
acrA基因在细菌多重耐药中起重要作用。利用PCR扩增副猪嗜血杆菌15个血清型标准菌株acrA基因,应用ClustalW进行氨基酸进化距离分析和同源性分析,绘制进化树。序列测定与分析表明,aC—rA基因在副猪嗜血杆菌15个标准菌株中都存在,其中,血清5型、8型、14型有2个拷贝(acrA1和acrA2)。进化距离分析表明不同血清型之间AcrA1氨基酸序列有较大的差异,同源性为89.8%~100%。同一血清型副猪嗜血杆菌aerA1和acrA2氨基酸同源性低于30%,但都位于比较保守的基因座上,这是首次发现副猪嗜血杆菌会存在2个acrA基因。本研究为副猪嗜血杆菌多重耐药机制的研究提供了理论基础,同时对新型抗菌药物的研发也具有重要的参考价值。
The acrA gene palys an importmant role in bacterial multidrug reisitance. To investigate the mo- lecular mechanisms of H. parasuis drug resistance, acrA gene was amplified from 15 reference strains of H. parasuis. Then, the amino acid phylogenetic tree and evolutionary distance analysis as well as se- quence homelogy analysis were performed using Clustal W. The sequence analysis indicated that acrA gene existed in 15 reference strains, interestingly, serovars 4, 8 and 14 of H. parasuis contain two copies of ac- rA ( acrA1 and acrA2). The evolutionary distance analysis showed a great difference from 15 serovars in AcrA1 amino acid sequences, the homology is 89.8% to 100% . The amino acid sequence similiarties of acrA1 and acrA2 in the same serovar of H. parasuis were less than 30~ ,but both genes were located in the conservative locus. It showed for the first time, to our knowledge, that certain H. parasuis strains contain two copies of acrA. The results of the present study provided foundation for further studies of mul- tidrug resistance of H. parasuis, and also had an importmant reference value for developing a new class of antimicrobial agents.
