软肝化坚颗粒药物血清对肝星状细胞信号传导通路的调节作用 被引量:4

摘要 目的:研究软肝化坚颗粒药物血清对活化的肝星状细胞(HSC)信号传导通路的调节作用。方法:通过灌胃制备小鼠软肝化坚颗粒及秋水仙碱药物血清。将HSC-T6细胞分为5组,分别加入含5%、10%、20%软肝化坚颗粒血清,10%秋水仙碱血清和10%正常小鼠血清的培养基,培养24小时后,收集各组细胞。采用荧光定量RT-PCR检测Ⅰ型大麻素受体mRNA(CB1 mRNA)、黏着斑激酶mRNA(FAK mRNA)和细胞外信号调节激酶mRNA(ERKmRNA)水平。结果:与正常血清组相比,秋水仙碱组、5%、10%及20%软肝化坚颗粒血清组细胞中CB1 mRNA的含量均明显降低(P值均为0.000;与秋水仙碱组相比,10%、20%软肝化坚颗粒血清组细胞中CB1 mRNA的含量均明显降低(P值均为0.000)。与5%软肝化坚颗粒血清组相比,10%、20%软肝化坚颗粒血清组细胞中CB1 mRNA的含量均明显降低(P值均为0.000);与10%软肝化坚颗粒血清组相比,20%软肝化坚颗粒血清组细胞中CB1 mRNA的含量均明显降低(P值为0.000)。与正常血清组相比,秋水仙碱组、5%、10%及20%软肝化坚颗粒血清组细胞中ERK mRNA的含量均明显降低(P值分别为0.000、0.002、0.000和0.000);与5%软肝化坚颗粒血清组相比,20%软肝化坚颗粒血清组细胞中ERK mRNA的含量明显降低(P值为0.014)。与正常血清组相比,秋水仙碱组、5%、10%及20%软肝化坚颗粒血清组细胞中FAK mRNA的含量均明显降低(P值分别为0.001、0.000、0.000和0.000)。结论:软肝化坚颗粒可能通过ERK通路和PI3K-Akt通路抑制HSC-T6细胞活化,发挥防治肝纤维化的作用。 Objective: To study the influence of Ruangan Huajian granule on transforming signaling in the activation of he- patic stellate cells. Methods: Mouse medicated serum was prepared with Ruangan Huajian granule by gastrogavage. HSC-6 were divided into five groups: control group incubated with normal mouse serum (10%) ; Ruangan Huajian granule group incubated with Ruangan Huajian granule mouse medicated serum (5% , 10% , 20% ) ; colchicine group incubated with colchicine mouse medicated serum ( 10% ) . Ceils of each group were cultured for 24 hours, and cells were collected. The mRNA levels of CB1, FAK and ERK were determined by quantitative RT-PCR. Results: The levels of CB1 mRNA in colchicine group and Ruangan Huajian granule group (5% , 10%, 20% ) were notably lower than those in control group, P value was 0. 000, 0. 000, 0. 000 and 0. 000, respectively. The levels of CB1 mRNA in Ruangan Huajian granule group ( 10%, 20% ) were notably lower than those in colchicine group, P value was O. 000 and 0. 000, respectively. The levels of CB1 mRNA in Ruangan Huajian granule group ( 10% , 20% ) were notably lower than those in Ruangan Huajian granule group (5%), P value was 0. 000 and 0. 000, respectively. The levels of CB1 mRNA in Ruangan Hnajian granule group (20%) were notably lower than those in Ruangan Huajian granule group ( 10% ), P value was 0. 000. The levels of ERK mRNA in colchicine group and Ruangan Huajian granule group (5%, 10%, 20% ) were notably lower than those in control group, P value was 0. 000 , 0. 002, 0. 000 and 0. 000, re- spectively. The levels of ERK mRNA in Ruangan Huajian granule group (20%) were notably lower than those in Ruangan Hua- jian Particle group (5%), P value was 0. 014. The levels of FAK mRNA in colchicine group and Ruangan Huajian granule group (5%, 10%, 20% ) were notably lower than those in control group, P value was 0. 001, 0. 000, 0. 000 and 0. 000, re- spectively.
