摘要
目的 探讨肺血管内巨噬细胞 (PIM)释放的细胞因子在感染性急性肺损伤 (ALI)发病中的作用。方法 仿Morton法灌洗肺血管床 ,贴壁法分离猪PIM ,培养于RPMI 16 40培养基 ,予 10 μg/ml脂多糖 (LPS)刺激 ,胸腺细胞增殖法测PIM培养上清白细胞介素 1β(IL - 1β)活性 ,酶联免疫吸附试验 (ELISA)法测肿瘤坏死因子α(TNFα)、白细胞介素 6 (IL - 6 )、白细胞介素 8(IL - 8)含量。结果 LPS刺激后 ,PIM释放TNFα、IL - 1β和IL - 6均呈单峰样显著增加 ,峰值分别出现在刺激后的 1h、2h和 4h(P <0 0 1) ;而IL - 8则于刺激后的 4~ 8h持续升高 (P <0 0 5~ 0 0 1)。结论 LPS刺激后 ,PIM分泌多种细胞因子 ,其中TNFα、IL - 1β升高最早 ,提示其在ALI的发生中起重要作用 ;而IL - 6、IL - 8升高较晚 ,且后者持续时间长 ,可能对ALI的病情进展起重要作用。
Objective To study the effects of cytokines released by pulmonary intravascular macrophage (PIM) on the infective acute lung injury (ALI).Methods Porcine pulmonary blood vessels were perfused by modified Morton's method.PIMs isolated with adhesion method were incubated in RPMI 1640 medium,and stimulated with lipopolysaccharide (LPS,10 μg/ml).The activity of interleukin (IL)-1 β and content of IL-6、IL-8 and tumor necrosis factor-α (TNFα) in the culture supernatants were measured by method of thymocyte proliferation or ELISA,respectively.Results After LPS stimulation,the release of TNFα,IL-1 β and IL-6 increased in a single peak manner ( P <0 01),and reaching their peak at 1 h、2 h and 4 h of poststimulation of LPS,respectively;however,continous increase of IL-8 was found from 4 h to 8 h after stimulation of LPS ( P <0 05~0 01).Conclusions Among the cytokines released by PIMs stimulated with LPS,the increase of TNFα and IL-1 β occurred earlier in comparison with IL-6 and IL-8,which suggested that the former take a part in the onset of ALI;whereas,IL-6 and IL-8 may be associated with the development of ALI.It also suggested that interaction of these cytokines is more important in the pathogenesis of ALI.
