六味地黄汤对5/6肾切除大鼠肌酐、尿素氮及24h尿蛋白定量的影响 被引量:3

Effects of Liuwei Dihuang decoction on creatinine,urea nitrogen,and 24-h urinary protein excretion in 5/6 nephrectomized rats

摘要 目的:观察六味地黄汤对5/6肾切除大鼠残肾的保护作用。方法:按照随机数字表法将72只SD雄性大鼠分为正常组(12只)、假手术组(12只)和手术组(48只)。手术组行5/6肾切除术诱导大鼠肾衰模型,假手术组同期行两次手术,每次仅打开腹腔,暴露肾脏后,再将肾脏纳入腹中,缝合关腹。造模后3d,将造模成功的43只大鼠随机分为模型组(15只)、依那普利组(14只)和六味地黄汤组(14只),分别对各组大鼠进行干预灌胃8周。观察大鼠毛色、行为及死亡率;所有大鼠每两周固定时间称重,留取24h尿,检验24h尿蛋白定量;于第8周抽取腹主动脉血,留取血清,检测血肌酐(SCr)、尿素氮(BUN)。结果:六味地黄汤组大鼠体质量增长情况接近正常组;六味地黄汤长期使用可降低5/6肾切除大鼠尿蛋白和SCr水平,对BUN水平影响不大。结论:六味地黄汤能有效保护5/6肾切除大鼠残余肾功能,降低血肌酐水平,改善大鼠生存质量。 Objective:To observe the protective effect of Liuwei Dihuang decoction on the remnant kidney in 5/6 nephrectomized rats. Methods : Seventy - two male Sprague - Dawley rats were divided into normal group ( n = 12 ), sham - operation group ( n = 12 ), and operation group( n = 48 ). The operation group underwent 5/6 nephrectomy to establish a rat model of renal failure. Each rat in the sham - operation group underwent operation twice in the same period;in each operation, the abdominal cavity was opened to expose the kid- neys, then the kidneys were put back into the abdominal cavity, and the abdominal cavity was closed by suture. At 3 days after the model was established ,43 rats among which the model was successfully established were randomly divided into model group (n = 15 ), enalapril group( n = 14) ,and Liuwei Dihuang decoction group( n = 14) to receive respective treatments by gavage for 8 weeks. The hair color,be- havior, and mortality were observed and calculated. Each rat was weighed every two weeks. The 24 - h urine was collected, and 24 - h u- rinary protein was quantified. Blood was collected from the abdominal aorta in the 8th week, and serum was obtained for measurement of serum creatinine (SCr) and blood urea nitrogen (BUN). Results:The Liuwei Dihuang decoction group had a body weight increase simi- lar to that of normal group. Long - term use of Liuwei Dihuang decoction decreased urinary protein and SCr levels in 5/6 nephrecto- mized rats,but it had little impact on BUN level. Conclusion:Liuwei Dihuang decoction can effectively protect the function of remnant kidney, decrease SCr level, and improve the quality of life in 5/6 nephreetomized rats.
