摘要
目的:研究抑睾调经汤对多囊卵巢综合征模型大鼠排卵功能障碍的干预作用,为健脾化痰,活血化瘀法应用于PCOS的治疗提供科学依据。方法:将雌性SD大鼠60只随机分为正常对照组、模型对照组、达英-35对照药组、抑睾调经汤高剂量治疗组、抑睾调经汤低剂量治疗组五组各12只,采用Poresky L法,皮下注射绒毛膜促性腺激素(HCG)、中性胰岛素,制备出PCOS模型大鼠。各组动物等容灌胃,正常对照组和模型对照组给予生理盐水,达英-35组、抑睾调经汤高、低剂量分别灌胃14 d,日2次。观察大鼠胰岛素(INS)、黄体生成激素(LH)、卵泡刺激素(FSH)、卵巢病理组织学的变化。结果:①抑睾调经汤组可显著降低PCOS模型大鼠血清LH水平,与模型组比较差异具有统计学意义(P<0.05);并能升高FSH,与模型组比较差异有统计学意义。②抑睾调经汤组能明显降低PCOS模型大鼠血清INS水平,与模型组比较差异有统计学意义(P<0.05)。③抑睾调经汤组能使PCOS模型大鼠恢复排卵,增加卵泡壁颗粒细胞层数和黄体组织数量。结论:抑睾调经汤具有调整下丘脑—垂体—卵巢轴,降低胰岛素水平,恢复排卵的作用。
Objective： To study the intervention effect of Yigao Tiaojing Decoction（YGTJD） on the ovulatory dysfunction of model rats with polycystic ovary syndrome（PCOS）,so as to provide scientific basis for the application of the method of strengthening the spleen and removing the phlegm,promoting blood circulation and removing blood stasis in the treatment of PCOS.Methods： 60 female SD rats were randomly divided into five groups： normal control group,model control group,Diane-35 control group,YGTJD high-dose group,YGTJD low-dose group,with 12 rats in each group.With Poresky L method,PCOS model rats were prepared by subcutaneous injection of human chorionic gonadotropin（HCG） and neutral insulin.Rats in each group were given isovolumetric gastric perfusion,the normal control group and the model control group received normal saline,while Diane-35 control group,YGTJD high-dose group and YGTJD low-dose group were given gastric perfusion 14 d,2 times a day.Observed the changes of insulin（INS）,luteinizing hormone（LH）,follicle stimulating hormone（FSH）,ovarian histopathology.Results： YGTJD groups could significantly decrease the level of serum LH of PCOS model rats,with significant statistical differences（P0.05） compared with that of the model group,and increased FSH,with significant difference compared with that of the model group.YGTJD groups could obviously reduce the level of serum INS of PCOS model rats,with the statistically significant difference（P0.05） compared with that of the model group.YGTJD could make PCOS model rats recover the ovulation,and could increase stratum granulose of ovarian follicle and quantity of luteum.Conclusion： YGTJD has the effects of adjusting hypothalamic-pituitary-ovarian axis,reducing insulin levels and recovering ovulation.
