摘要
为研究银鲳幼鱼在不同盐度环境中的耐受状况,选取平均质量为(1.004±0.188)g的银鲳幼鱼作为试验对象,设置12、20和28三个盐度梯度,分别在0 d、0.5 d、1 d、2 d、4 d、8 d取样,检测皮质醇、热休克蛋白-70、溶菌酶、碱性磷酸酶、Na+-K+-ATP酶活性这些应激和免疫指标的变化,并记录各盐度下银鲳幼鱼的存活率。试验结果表明,银鲳幼鱼在三个盐度环境中养殖8 d的存活率无显著性差异。应激指标的结果显示,皮质醇比热休克蛋白更能体现机体的应激程度,银鲳幼鱼经8 d恢复至正常水平,盐度12环境中应激程度较其他两个盐度强,但到第8 d三个盐度下并无显著差异。银鲳幼鱼在养殖过程中溶菌酶、碱性磷酸酶、Na+-K+-ATP酶活性呈现不同的变化趋势,盐度变化对溶菌酶、Na+-K+-ATP酶活性影响较大,至第8 d各盐度下均无显著性差异。因此,银鲳幼鱼在不同盐度环境中养殖应激程度和免疫水平不同,至第8 d三个盐度下均无显著差异,表明银鲳幼鱼对低盐的适应能力较强。
In order to investigate the tolerance of juvenile fish,the Pampus argenteus juveniles were cultured at water salinities of 12,20,28,for eight days.Cortisol,heat shock protein-70,lysozyme,alkalin phosphatase,Na+-K+-ATPase activity,and survival rate were determined at 0 d,0.5 d,1 d,2 d,4 d and 8 d,respectively.The results showed that,the survival rates of P.argenteus at the three salinities for 8 days had no significant difference.From the results of stress indicator,cortisol was more sensitive to environmental change than heat shock protein.The juvenile P.argenteus recovered to normal condition over 8 days,and the stress extent in salinity of 12 was stronger compared to other salinities.During the stress process,lysozyme,alkaline phosphatase and Na+-K+-ATPase activity displayed different changes,and the effects of salinity on lysozyme and Na+-K+-ATPase activity were stronger.There were no significant differences of all parameters at the eighth day among different salinities.Therefore,the juvenile P.argenteus acclimatized in different salinities showed different immune responses and adaptation,and returned to normal physiological condition until the eighth day,suggesting a strong adaptability of juvenile P.argenteus to low salinity.
