运动对Apo E基因敲除小鼠冠状动脉超氧化物阴离子和NF-κB的调控作用 被引量:1

Regulatory effect of exercise on the expressions of superoxide anion and nuclear factor in coronary artery of apolipoprotein E-deficient mice

摘要 目的:探讨长期中等负荷耐力运动对载脂蛋白E(ApoE)基因敲除小鼠冠状动脉粥样硬化的影响和对冠状动脉超氧化物阴离子、CD40L的调控作用。方法:40只6周龄ApoE基因敲除小鼠随机分为运动干预组和对照组。运动干预组给予长期中等负荷耐力跑台运动干预32周;一般对照组未予以特殊干预。实验结束后评估两组小鼠冠状动脉主干的动脉粥样硬化病理形态学改变程度,采用荧光免疫组化法测定冠状动脉主干的超氧化物阴离子、CD40L的表达、实时PCR的方法测定NF-κB、MCP-1mRNA的表达。结果:一般对照组冠状动脉粥样硬化进展显著;长期中等负荷耐力运动组冠状动脉粥样硬化程度显著下降、冠状动脉主干狭窄率较对照组显著下降(33.3±5.66%vs48.3±5.91%,P<0.01)、冠状动脉主干超氧化物阴离子显著低于对照组(2.17±0.69vs3.70±0.51,P<0.01),CD40L的表达均显著低于对照组(1.97±0.41vs2.47±0.35,P<0.01),并且NF-κB、ICAM-1、MCP-1mRNA的表达也显著低于对照组。结论:长期中等负荷耐力运动可显著下调ApoE基因敲除小鼠冠状动脉超氧化物阴离子与CD40L的表达,减轻炎症浸润并延缓冠状动脉粥样硬化的进展。 Objeetlve:To investigate the regulatory effect of exercise on the expressions of superoxide anion and inflamma- tory reaction changes in coronary artery of apolipoprotein E(Apo E) deficient mice and the atherosclerotic pro- gression of main coronary artery by treating with long-term medium load endurance exercise. Method: Forty 6-week-age Apo E deficient mice were randomly divided into two groups: the exercise group and the control group (n=20, each). The mice in the exercise group were kept on a long-term of 32 weeks treadmill exercise. At the end of study, the morphology changes of main coronary artery were evaluated, the expressions of superoxide anion and leukocytel differentiation antigen 40 ligands (CD40L) on the coronary ar- tery were measured by fluoroimmanohistochemical technique, and the expressions of inter cellular adhesion mol- ecule(ICAM)-l, monocyte chemoattractant protein(MCP)-i and NF-κB were also measured by real-time fluores- cence quantitative PCR technique.Result: The atherosclerotic stenosis severity of main coronary artery in the control group was more seriously than that of the exercise group (48.3±5.91% vs 33.3+5.66, P〈0.01). The expressions of superoxide anion and CD40L on coronary artery were significantly lower in the exercise group than that of the control group(2.17± 0.69 vs 3.70:e0.51, P〈0.01; 1.97±0.41 vs 2.47±0.35, P〈0.01). The mRNA expressions of ICAM-1, MCP-1 and NF-r,B were also significantly lower in the exercise group than that of the control group. Conclusion: Long-term medium load endurance exercise can inhibit the superoxide anion expression, reduce the inflammation reaction on coronary artery and slow down the progression of coronary artery atherosclerosis of Apo E deficient mice.
