丹参酮ⅡA对压力超负荷心肌肥厚大鼠交感神经重构的影响 被引量:2

Effect of Tanshinone ⅡA on the Sympathetic Reconstruction in Rats with Cardiac Hypertrophy Induced by Pressure Overload

摘要 目的丹参酮ⅡA对压力超负荷心肌肥厚大鼠交感神经重构的影响。方法将40只大鼠随机分为模型组(TAC)和假手术组(Sham),每组均随机给予丹参酮ⅡA(TanⅡA)或生理盐水(Saline)处理,观察连续治疗12周后的左心室的生长相关蛋白43(GAP43)、酪氨酸羟化酶(TH)蛋白、神经生长因子(NGF)表达水平、GAP43和TH阳性神经纤维密度。结果与Sham+Saline组相比,TAC处理可导致左心室的TH、GAP43及NGF的蛋白水平上升,且GAP43和TH阳性神经纤维密度均上升;Tan ⅡA可有效地改善TAC的症状;TanⅡA对Sham组的各项指标没有影响。结论压力超负荷心肌肥厚大鼠的交感神经发生重构,采用TanⅡA可改善心肌交感神经重构。 Objective:To explore the effect of Tanshinone Ⅱ A on the sympathetic reconstruction in rats with cardiac hypertrophy induced by pressure overload.Methods:40 rats were randomly divided into Model Group and Sham Group in which Tanshinone ⅡA(Tan ⅡA) or Saline were employed.Twelve weeks later,the following indices were measured such as the protein level of growth associated protein 43(GAP43),tyrosine hydroxylase(TH) protein and nerve growth factor(NGF) in left ventricular as well as the GAP43 and TH positive nerve fiber density.Results:Compared with Sham +Saline group,TAC treatment could result in the elevated expression of NGF,TH and GAP43 protein levels in LV,and the increased GAP43 and TH positive nerve fiber density;Tan Ⅱ A could effectively improve the symptoms of TAC;Tan ⅡA did not alter the indicators of the Sham group.Conclusion:Pressure overload induce cardiac sympathetic reconstruction,which can be improved and attenuated by Tan ⅡA treatment.
