摘要
目的探讨温通法对S180荷瘤鼠的抑瘤机制,为其临床应用提供理论依据。方法建立昆明种小鼠S180肉瘤模型50个。随机分为中药高剂量组、中药中剂量组、中药低剂量组、荷瘤对照组、CTX化疗组。通过SP免疫组化法检测S180肿瘤细胞各组Bcl-2、P53基因蛋白表达情况。结果 Bcl-2蛋白和P53蛋白在荷瘤对照组与中药中剂量组、中药高剂量组、CTX组的表达比较均有显著差异,其中与中药中剂量组比较有统计学意义(P<0.05),与中药高剂量组、CTX化疗组比较有显著统计学意义(P<0.01);中药高剂量组与CTX化疗组比较均无显著性差异(P>0.05)。结论温通法抑瘤机制可能是通过抑制P53基因突变,下调Bcl-2蛋白的表达水平实现的。
Objective Exploring WenTong method to tumor-burdened rat S180unti-tumor mechanism,in order to provide the theory basis for its clinical application.Method:Making 50 the Kunming-mice S180sarcoma models.They were randomly divided into high,middle,low dose group traditional Chinese medicine group,a tumor-burdened control group,CTX chemotherapy group.Detecting S180 tumor cell in Bcl-2,P53 protein gene expression by through the SP immunohistochemical method.Results:Compared to high,middle dose group traditional Chinese medicine group,CTX chemotherapy group,the tumor-burdened control group had improvince in Bcl-2 protein gene expression(P〈0.05).Compared to high dose group traditional Chinese medicine group,CTX chemotherapy group,the tumor-burdened control group had signifikently improvince(P〈0.01).High dose group traditional Chinese medicine group and CTX chemotherapy group have no difference(P〈0.05).Conclusion:The unti-tumor mechanism may be that WenTong method can inhibiting P53 gene mutations,cut down Bcl-2 protein expression level.
