香蕉皮黑色素提取工艺研究 被引量:5

摘要 以香蕉皮为原料,以一定浓度的香蕉皮黑色素溶液在410nm处的吸光度值为指标,通过正交实验,对碱-酸法、超声波辅助提取法、酶-超声双辅助提取法3种不同提取方法的效果进行了比较,以期优化香蕉皮黑色素的提取工艺,为香蕉皮黑色素提取工业化生产提供理论指导。结果表明:碱-酸法最佳条件为盐酸浸泡时间为0h,提取时间为20min,加碱后溶液pH=14,提取温度为70℃,吸光度值为0.295A;超声波辅助提取法最佳条件为超声时间为10min,超声功率为100W,加碱后溶液pH=14,盐酸浸泡时间为0h,提取温度为70℃,吸光度值为0.573A;酶-超声双辅助提取法最佳条件为纤维素酶与果胶酶协同作用,酶的作用时间为1.5h,酶作用环境为pH=4,加碱后溶液pH=14,超声功率为100W,超声时间为10min,盐酸浸泡时间为0h,提取温度为70℃,吸光度值为0.923A。可见,酶-超声双辅助提取法是香蕉皮黑色素实现工业化生产较为理想的生产工艺。 Taking banana skin as raw material,with the absorbance at 410 nm as the index, the base-acid method, ultrasonic assisted extraction and enzyme-ultrasonic double auxiliary extraction was discussed,which was more fit for melanin, by the orthogonal test,in order to optimize the extraction technology of melanin from banana skin, and provide theoretical guidance for industrialization production. The results showed that base-acid method optimum conditions were hydrochloric acid soaking time 0 h, extracting time 20 min, alkaline solution pH 14, extraction temperature 70℃, absorbance value 0. 295 A; ultrasonic assisted extraction optimum conditions were ultrasonic time 10 min,ultrasonic power 100 W,alkaline solution pH= 14, hydrochloric acid soaking time 0 h,extracting time 20 min, extraction temperature 70℃, absorbance value 0. 573 A;enzyme-ultrasonic double auxiliary extraction optimum conditions were cellulase and pectinase synergy, enzyme action time 1.5 h, enzyme action environment pH= 4, alkaline solution pH= 14, ultrasonic power 100 W, ultrasonic time 10 rnin, hydrochloric acid soaking time 0 h, extracting time 20 min, extraction temperature 70℃, absorbance value 0. 923 A. In conclusion,enzyme-ultrasonic double auxiliary extraction was ideal industrialize production process for melanin extracted form banana skin,
