摘要
目的:比较桑叶不同有效部位对2型糖尿病大鼠的治疗作用。方法:SD大鼠随机分为正常对照组(NC组8只),糖尿病组。糖尿病组大鼠利用高脂高糖饲料喂养8周联合一次性腹腔注射STZ 30mg.kg-1建立2型糖尿病大鼠模型,造模成功的大鼠随机分为模型对照组(DM组),桑叶黄酮组治疗组(MTF组,30mg.kg-1),桑叶多糖组治疗组(MTP组,100mg.kg-1),桑叶水提物治疗组(MWE组,6g.kg-1)和罗格列酮治疗组(RSG组,4mg.kg-1),每组10只。灌胃给药,每周测空腹血糖FBG,于第6周各组大鼠行灌胃葡萄糖耐量(oralglucose tolerance test OGTT)试验,测定大鼠血清胰岛素(insulin INS)、游离脂肪酸(free fatty acids NEFA)、胆固醇(cholesterol CHO)、三酰甘油(triglyceride TC)、丙二醛(malonaldehyde MDA)含量;取胰岛组织,光镜下观察胰岛组织病理学形态变化。结果:桑叶各个有效部位治疗后,各组大鼠FBG、血清TG、CHO、MDA均显著下降,INS显著升高,OGTT试验糖耐量水平显著增高,胰岛病理损伤减轻。结论:桑叶各个有效部位都具有显著的降低空腹血糖,改善糖耐量的作用,且在降低血糖的同时降低血脂,保护胰岛细胞。桑叶各个不同有效部位在降血糖、改善血脂方面的作用表现不一。
OBJECTIVE To investgate the therapeutic effects of different effective extracts of Morus alba L.leaf on type 2 diabetes mellitus rats(T2DM rats).METHODS SD rats were randomly divided into normal control group(group NC,n=8) and diabetes mellitus group(DM),the T2DM rats model were induced by feeding with high-sugar and high-lipid fodder for 8 weeks and a single intraperitoneal injection of streptozotocin(STZ),the successful models were divided into 5 groups at random: diabetes mellitus model group(DM),diabetes with mulberry leaves flavone extract(30 mg·kg^-1 treatment group(MTF),diabetes with mulberry leaves polysaccharide extract(100 mg·kg^-1 treatment group(MTP),diabetes with mulberry leaves aqueous extract(6 g·kg^-1 treatment group(MEW),diabetes with rosiglitazone(4 mg·kg^-1 treatment group(RSG),10 rats per group.The drug was administered by intragastric administration and the rats were measured the fasting plasma glucose level weekly,additionally,the glucose tolerance factor(OGTT assay) was evaluated in the 6th week,rats blood plasma insulin(INS),free fatty acids(FFA),cholesterol(CHO),triglyceride(TC) and malonaldehyde(MDA) levels were measured,the function of islets,fat levels in the blood and anti-oxidative ability were evaluated accordingly,finally,taking islet tissue to observe the changes in morphology and pathology under a light microscope.RESULTS After six weeks treatment of different effective extracts of mulberry leaf,the rats FBG,blood plasma TG,CHO,MDA significantly decreased,blood plasma insulin significantly increased and so did the oral glucose tolerance test level.Meanwhile,different effective extracts of mulberry leaf also markedly attenuated the pathological injury in pancreas.CONCLUSION Each effective extract of Morus alba L.leaf could reduce fasting blood glucose and improve glucose tolerance significantly,meanwhile,they had the effects of decreasing blood glucose,reducing the blood lipid and protecting human pancreata
