摘要
目的观察华蟾素注射液对人肝癌HepG-2细胞增殖、凋亡及拓扑异构酶Ⅱ表达的影响。方法分为对照组与实验组(3个剂量0.10,0.21,0.42μg·mL-1华蟾素注射液);用MTT法观察华蟾素对HepG-2细胞增殖的影响;用流式细胞术法检测华蟾素对HepG-2细胞周期的影响;用逆转录—多聚酶连反应法检测华蟾素对肝癌HepG-2细胞拓扑异构酶Ⅱ(TOPO酶Ⅱ)的mRNA水平表达的影响;用蛋白免疫印迹法观察华蟾素对HepG-2细胞中TopoⅡ蛋白表达的影响。结果华蟾素注射液对HepG-2细胞增殖具有抑制作用,呈时间和剂量依赖性;阻滞HepG-2细胞于S期,可明显下调TopoⅡmRNA(P<0.05)及蛋白的表达(P<0.05),均呈剂量依赖性。结论华蟾素注射液能够抑制人肝癌HepG-2细胞增殖,下调TOPOⅡ的表达是其可能的药效之一。
Objective To investigate the effects of Cinobufacini injections on the expression of topoisomeraseⅡof hepatocarcinoma HepG-2 cells.Methods Experiment were divided into control group and test group(the different Cinobufacini injections group 0.10,0.21,0.42 μg.mL-1).Cell proliferation was assessed by MTT assay.Cell cycles were shown through FCM.The expression of topoisomerase(TOPO Ⅱ) mRNA were analyzed by RT-PCR.Expressions of TOPO Ⅱprotein were analyzed by Western-Blot.Results Cinobufacini injection significantly inhibited HepG-2 cell proliferation(P 0.05) in dose and time-dependent ways,induced tumor cells arrested at the S-phase a dose dependent manner.Down regulated mRNA and protein expressions of TOPOⅡ(P 0.05) in a dose dependent manner.Conclusion Cinobufacini injections can inhibit human hepatocellular carcinoma HepG-2 cells proliferation,induced tumor cells arrested at the S-phase,regulation of TOPOⅡexpressions might be one of the mechanisms of Cinobufacini injections anti-tumor.
