摘要
目的 :建立毛细管气相色谱 电子捕获检测法 (GC ECD)测定人血浆中 5 单硝酸异山梨酯浓度的方法。方法 :以对硝基苯乙腈为内标 ,血样经乙醚 :正己烷 (4∶1)一次提取后 ,采用HP 5 0 +毛细管柱测定。结果 :检测浓度在 10~ 10 0 0ng·ml-1内 ,回归方程为Y =0 .0 138+ 1.35 76× 10 -3 X ,r=0 .9997。血浆中最低检测浓度为 5ng·ml-1。平均回收率为 10 0 .80 % ,日内、日间RSD均小于 3%。结论 :该法处理血样简单 ,测定结果准确度高 ,专一性强 ,适用于药动学的研究。
OBJECTIVE:To establish a GC-ECD method for determination of isosorbide-5-mononitrate in human plasma.METHODS:Using 4-nitro-phenylacetonitrile as internal standard.Plasma samples were extracted with a solvent of ether and n -hexane(4∶1) ,and determined with HP-50+capillary column.RESULTS:The linear regressin equation was Y = 0.0138 + 1.3576 ×10 -3 X , r = 0.999 7 in the concentration range from 10 to 1000 ng·ml -1 .Detection limit was 5 ng·ml -1 .The average recovery was 100.80% .The RSD % was less than 3%.CONCLUSIONS:The method was simple,precise,and suitable for pharmacokinetic studies in human beings.
