摘要
目的探讨局灶性脑缺血大鼠梗死灶周围Nogo-A受体(NgR)的表达及细胞内定位。方法电凝右侧大脑中动脉建立大鼠局灶性脑缺血模型,免疫组织化学染色检测脑缺血后第5、14、28天脑缺血大鼠梗死灶周围皮层NgR表达及其细胞内定位,并与假手术大鼠相应部位皮层NgR表达作比较。结果脑缺血大鼠梗死灶周围皮层NgR表达第5天低于假手术大鼠,在第14、28天明显高于假手术大鼠。在假手术大鼠皮层、脑缺血大鼠皮层第5天NgR表达主要表达在神经胞体,脑缺血大鼠第14、28天皮层NgR轴突远端的表达增强。结论梗死灶周围皮层NgR表达在缺血后的不同时期表达量及细胞内定位不同,可能与其功能有关。
Objective To investigate the dynamic expression and cellular localization of Nogo-A receptor(NgR) in the perilesional cortex following focal cerebral ischemia in rats.Methods The focal cerebral ischemia model was established by electrocoagulating the right middle cerebral artery in rats.The expression and location of NgR in the perilesional cortex.Methods The focal cerebral ischemia model was established by electrocoagulating the right middle cerebral artery in rats.The expression and location of NgR in the perilesional cortex were assessed by immunohistochemical staining.Results The expression of NgR in the perilesional cortex was down-regulated on the 5th day after cerebral ischemia comparing with sham operation group;however,up-regulated on the 14th day and the 28th day.Prominent cell body expression of NgR was observed in cortex neuron of sham operation group and perilesional cortex neuron of ischemia group on the 5th day;while superseded axon expression was observed in perilesional cortex neuron on the 14th day and 28th day.Conclusion The NgR expression in the perilesional cortex differed during different periods,indicating its different function in cerebral ischemia.
