摘要
目的:研究伏毛铁棒锤活性部位化学成分及抗炎镇痛作用。方法:通过二甲苯致小鼠耳廓肿胀法、鸡蛋清致小鼠足肿胀法、小鼠醋酸扭体法和小鼠热板法,研究伏毛铁棒锤大孔树脂法富集的30%乙醇部位的抗炎镇痛作用。通过硅胶柱色谱、ODS反相柱色谱、凝胶柱色谱等多种分离手段,对30%乙醇部位进行系统分离,利用1H-NMR、13C-NMR、DEPT等波谱技术进行单体化合物结构解析。结果:小鼠耳廓肿胀实验表明30%乙醇部位组与模型对照组相比有显著性差异(P<0.05),小鼠足肿胀反应的实验显示30%乙醇部位组与模型对照组相比有极显著性差异(P<0.01)。小鼠醋酸扭体实验结果显示30%乙醇部位组与模型对照组相比有显著性差异(P<0.05),小鼠热板实验结果表明30%乙醇部位组与模型对照组相比,在给药30和45 min后有显著性差异(P<0.05),在60min后无显著性差异(P>0.05)。从伏毛铁棒锤30%乙醇部位分离出14个单体化合物,目前鉴定出7个化合物,分别为:蔗糖(I)、蒙花苷(Ⅱ)、β-谷甾醇(Ⅲ)、次乌头碱(Ⅳ)、乌头碱(Ⅴ)、β-胡萝卜苷(Ⅵ)和3,5,6-三羟基-β-谷甾醇(Ⅶ)。结论:伏毛铁棒锤30%乙醇部位具有抗炎镇痛的药理活性。化合物Ⅰ、Ⅱ、Ⅳ、Ⅵ、Ⅶ为首次从该植物中分离得到。
Objective : To study the chemical compositions of active site of Aconitum flavum and their pharmacological effects on anti-inflammatory andantalgic. Methods: Xylene-induced mice ear swollen and egg white-induce toe swollen test were used to assess the anti-inflammatory effect of Aconitumflavum. Acetic acid-induce mice writhing and hot-plate test were used to assess the antalgic effect of Aconitumflavum. Silica gel,ODS and Sephadex LH-20 techniques were employed for isolation and purifcation the constituents of Ac-onitumflavum. The chemical compounds were identified by ^1H-NMR, ^13 C-NMR, DEPT, HMBC, HMQC and COSY. Results: When com-pared with model, group,there was significant difference in 30% alcohol active extract group (P 〈0.05 ) in xylene-induce mice eax-swollen test. In the experiment of egg white-induce toe swollen test,30% alcohol active extract group had great significant difference when compared with the model group (P 〈 0.01 ) . In acetic acid writhing test ,30% alcohol active extract group had significant analge-sic action when compared with model group. In hot plate test,when compared with model group,the analgesic eftbct of 30% alcohol ac-tive extract group ( P 〈 0.05 ) at 30 and 45 minutes after administration was significanty increased. Fourteen compounds were isolated from the active extract ,while seven compounds were identified as: sucrose( Ⅰ ) ,linarin( Ⅱ ) ,β-sitosterol(Ⅲ ) ,.β-daucosterol (Ⅳ) , β-sitosterol-3,5,6 -triol ( Ⅷ ), aconitine ( Ⅴ ) and hypaconitine ( Ⅳ ). Conclusion : 30 % alcohol active extract of Aconitum flavum has anti-inflammatory and antalgic effects. Compounds Ⅰ , Ⅱ , Ⅳ, Ⅶ and Ⅷ are isolated for the first time.
