摘要
目的建立孕期丙戊酸钠注射致Sprague-Dawley子代大鼠孤独症的模型,探讨模型的神经行为学特点。方法随机选择孕12.5 d SD大鼠8只,随机分组,VPA组(4只)腹腔注射丙戌酸钠(VPA)(600 mg/kg),Con组(4只)腹腔注射等量的生理盐水。分别对两组的子代大鼠进行水迷宫、旷场试验和自发运动等神经行为学检测。结果 VPA组仔鼠病死率9%、畸形率4.6%,畸形仔鼠出现尾部弯折,Con组死亡1只、无畸形。与Con组相比较,VPA组仔鼠显示低体质量(P<0.05)、睁眼时间延长(P<0.05)、重复行为增多,修饰行为、群集性活动减少,攻击性行为增加。仔鼠生后45 d水迷宫VPA组结果显示:到达平台潜伏期及正确穿越平台象限次数均略少于Con组,但无明显差别。旷场试验中VPA组站立次数(13.50±10.65)次、跨格子数(48.420±4.037)个均显著低于Con组(26.125±9.701,91.571±4.080)个(P<0.05)。自发运动中,VPA组仔鼠表现出典型的活动过多或者活动过少的极端规律,活动过多组11只自发运动次数为(168.33±15.38)次,活动过少组5只自发运动次数为(31.85±3.66)次,与Con组自发运动次数(59.110±3.007)次比较,三组比较差异存在统计学意义(P<0.05)。结论丙戊酸钠孕中期注射可以导致子代Sprague-Dawley大鼠出现典型的孤独症症状,且出现典型的自发运动过多、过少等更为复杂的孤独症症状。
Objective To establish a rat model of autism induced by prenatal exposure to valproate and explore the neurobehavioral changes of newborn rats. Methods 8 pregnant SD rats (12.5d) were randomly divided into 2 groups. 4 pregnant rats in the experimental group were received a single intraperitoneal injection of 600 mg/kg sodium valproate (VPA) and rats in the control group were received phys- iological saline. Neurobehavioral influences of Offspring from two groups were detected by Morris water test, locomotors active test and open field test. Results The mortality and malformation rate in experimental group were 9% and 4.6% and no dead and no deformed were found in control group. Compared to control group, the offspring in experimental group showed lesser of the body mass ( P 〈 0.05 ), delayer eyes o- pening (P 〈 0.05 ), significantly increased locomotor and repetitive activities, lesser grooming behavior and group activities, and more at- tack. The offspring of rats in experimental group had less frequency of standing( 13.50 ± 10.65 ) and crossing cell (48.42±4.037 ) than con- trol group(26. 125±9. 701,91. 571±4.080)in open field test( P 〈 0.05 ). In Locomotor active test, 11 rats had more activities (168.33 ±15.38) and 5 rats had less activities (31.85±3.66), but the activities in control groups was (59.11±3. 007) , the difference among three groups is significantly (P 〈 0.05 ). Conclusion The results demonstrate that offspring of SD rats which prenatal rats" exposure to valproate have much more typical and complicated clinical symptoms of autism.
