白藜芦醇抑制辐射诱导的IL-1β表达 被引量:2

Effect of resveratrol on the expression of IL-1β in mesenchymal stem cells exposed to ionizing radiation

摘要 目的研究白藜芦醇对问充质干细胞(MSCs)受照后产生的炎症因子IL-1β的影响,探讨白藜芦醇对MSCs的辐射防护作用。方法将间充质于细胞分为空白对照组、单纯照射组与白藜芦醇组,白藜芦醇组预先给予不同浓度的白藜芦醇,经^137Csγ射线照射后,应用ELISA、Westernblot和RT—PCR等方法检测IL-1β的分泌和表达。结果预先给予间充质干细胞50、100和200μmol/L的白藜芦醇组与单纯照射组相比,IL-1β胞外分泌显著降低(t=83.34、24.48、12.52,P〈0.05),同时细胞内IL-1β蛋白表达和mRNA水平也显著降低(t=8.69、14.77、13.90,P〈0.05)。结论白藜芦醇可明显抑制辐射诱导的间充质干细胞炎症因子IL-1β的产生。 Objective To investigate the effect of resveratrol on IL-1β production in mesenchymal stem cells (MSCs) exposed to radiation and evaluate the radioprotective effect of resveratrol. Methods MSCs were divided into non-treated control group, irradiated group and resveratrnl-treated group where resveratrol with different dosage of 50, 100 and 200 p^mol/L was administrated to MSCs before irradiation. The expressions of IL-1β mRNA and protein were detected using real time PCR, ELISA, and Western blot, respectively. Results Compared to the irradiation group, pretreatment of cell with resveratrol reduced the external secretion of IL-1β ( t = 83.34, 24.48, 12. 52, P 〈 0.05) as well as the intracellular expression of IL-11β protein and mRNA ( t = 8.70,14.77,13.90, P 〈 0. 05 ). Conclusions Resveratrol could inhibit the induction of inflammatory factor IL-1β in the irradiated MSCs.
