转MAPK双链RNA干扰表达载体黄瓜对根际土壤真菌群落多样性的影响 被引量:5

摘要 为了明确MAPK双链RNA干扰表达载体转基因黄瓜植株对根际土壤真菌区系的影响,本研究采用18S rRNA基因克隆文库方法对转和非转基因黄瓜根际土壤真菌群落多样性进行分析,结果表明2种土壤的真菌类群包括子囊菌亚门真菌、担子菌亚门真菌、壶菌门真菌、未定地位真菌和未分类真菌,子囊菌亚门真菌为优势菌群。非转基因黄瓜土壤真菌多样性略高于转基因黄瓜土壤,但优势真菌梭孢壳菌Thielaviaaustraliensis的比例(34.7%)明显低于转基因黄瓜土壤(65.1%),而非转基因黄瓜土壤中比例较高的尖孢镰刀菌Fusarium oxysporum(12.9%)未在转基因黄瓜土壤中发现。这表明MAPK双链RNA干扰表达载体的转基因黄瓜植株对土壤真菌具有一定的影响,对土壤中某些类群真菌具有一定的抑制作用。 In order to evaluate the ecological safety of transgenic cucumber with double-strand RNA interference expression vector of MAPK, authors analyzed the diversity of fungal community from soil planted with non-transgenic and transgenic cucumbers with 18S rRNA gene clone library. Fungi in the two soils included Ascomycota, Basidiomycota, Chytridiomycot, incertae sedis fungi and unclassified fungi. The diversity of fungal community in non-transgenic cucumber soil was higher than that in transgenic cucumber soil. However, the dominant fungi Thielavia australiensis, in non-transgenic cucumber soil was markedly lower than that of transgenic cucumber soil (34.7% vs 65.1%). Moreover, Fusarium oxysporum was the predominant fungus in non-transgenic cucumber soil (12.9%) but not found in transgenic cucumber soil. These results reflected that the community of rhizosphere soil fungi was affected at some extent by transgenic cucumber, and some groups of fungi were inhibited by the transgenic cucumber.
