热休克蛋白70联合含CpG寡核苷酸的免疫治疗作用 被引量:1

The Immunotherapy Effect of the Combination of Tumor-Derived HSP70-Pcs and Cpg-ODN

摘要 目的探讨肿瘤热休克蛋白70(HSP70-PCs)联合含CpG寡脱氧核苷酸(CpG-ODN)对荷瘤模型的治疗作用及可能机制。方法纯化肝癌细胞中的HSP70-PCs,SDS-PAGE及Western-blot技术对产物进行鉴定。建立小鼠肝癌皮下移植瘤模型,分别运用HSP70-PCs、CpG-ODN和HSP70-PCs联合CpG-ODN对其进行治疗,观察治疗作用。ELISA进行免疫后细胞因子分析。结果二者联合可产生强大的抗肿瘤作用,肿瘤体积、质量、抑瘤率、肿瘤转移率及生存期限与两药单独使用比较差异有统计学意义;二者联合可以提高荷瘤小鼠血清IFN-γ及CXCL-10水平,与二者单独使用比较差异有统计学意义。结论联合使用HSP70-PCs和CpG-ODN能显著提高机体免疫功能,对荷瘤小鼠有显著治疗作用,可能与诱导Th1型细胞因子产生有关。 Objective To investigate the immunotherapy effect and mechanism of the combination of HSP70-PCs and CpG- ODN on tumor-bearing mice. Methods HSP70-PCs was purified from the hepatoma cell Hepa 1 - 6, then identified by SDS- PAGE and Western blot. The anti-tumor efficacies of mono-administration of HSP70-PCs or CpG-ODN and their combination were observed by animal experiment on mice hepatoma models. Serum IFN-T and CXCL-10 levels were detected by ELISA. Results The co-use of HSP70-PCs and CpG-ODN was more effective than either of the two therapeutic agents alone. The tumor volume, weight,inhibited rate, metastasia rate, and average life span of mice of co-use group were significant difference between the HSP70-PCs group and CpG-ODN group. The levels of serum IFN-T and CXCL-10 in co-use group were notably promoted than the other two agents administrated alone. Conclusion The combination of HSP70-PCs and CpG-ODN enhanced the immunologic competence of organism, and appeared significantly therapeutic effect on mice hepatoma model by enhencing the serum IFN-T and CXCL-10 levels.
