胰高血糖素样肽2(GLP-2)促进肠切除术后大鼠小肠黏膜增殖及其与时间的相关性 被引量:2

摘要 目的探索胰高血糖素样肽2(glucagon-like peptide-2,GLP-2)促进肠切除大鼠术后残余小肠黏膜的增殖情况及其与时间的相关性。方法 30只健康雄性SD大鼠随机分为实验组、对照组和空白组(每组10只)。实验组和对照组建立小肠切除术模型,实验组术后每日注射GLP-2、对照组和空白组注射等量生理盐水,分别在注射当天、术后第7天和第14天分批处死大鼠,随机选取残余肠黏膜行HE染色,并计算其肠绒毛高度、厚度及陷窝深度。结果实验组较对照组、对照组较空白组在肠绒毛高度、厚度、陷窝深度的数值均有明显升高(P<0.05),且实验组第14天较第7天时数值亦有明显升高(P<0.05)。结论 GLP-2能有效改善肠切除大鼠术后小肠的黏膜增殖功能,且随着应用时间延长效能增加,并在2周后达到术前水平。 Objective To investigate the effect of glucagon-like peptide-2 (GLP-2) on proliferation of intestinal mucosa in rats following small bowel resection and its time-dependent pattern. Methods Thirty healthy male rats were randomly assigned into case, control and blank group (10 for each group). The intestinal resection models were established on rats in case and control group. GLP-2 of 0.2 μg g-1 . d-1 was injected into rats in case group,meanwhile,the same volume of 0.9% saline was injected into control and blank group. The rats were sacrificed at day 0,7 and 14 after surgery. HE staining of mucosa from residual intestine was performed to observe the length, thickness and lacunae of intestinal villus. Results The length,thickness and lacunae of intestinal villus in case group were significantly increased comparing with control and blank group. Significant improvement was also observed in day 14 comparing with day 7 after surgery in case group. Conclusions GLP-2 can effectively improve the proliferation of intestinal mucosa in rats following small bowel resection in a time-dependent pattern,which indicated a fully recover at day 14 after surgery.
