摘要
目的观察阿尔茨海默病(AD)转基因小鼠不同脑区β-淀粉样蛋白(Aβ)与微管相关蛋白1轻链3(LC3)的分布,探讨二者的变化及其相关性。方法应用免疫组织化学和体视学方法检测Aβ与LC3在AD转基因小鼠各脑区的含量变化。结果 (1)突变型小鼠脑内的Aβ与LC3表达量多于野生型小鼠;(2)Aβ与LC3在突变型小鼠的M1、M2、GD、CA1、CA3、CPu的表达量高于野生型小鼠的相应区域;(3)Aβ与LC3在突变型小鼠海马的GD、CA1、CA3脑区与躯体运动区的M1、M2中的表达量相当,且上述区域的表达量多于CPu,在Cb中表达最少;(4)突变型小鼠LC3与Aβ的分布呈正相关性(r=0.486,P<0.05)。结论 (1)Aβ与LC3在野生型与突变型小鼠相应脑区的分布具有差异性,且二者在突变型小鼠不同脑区中的表达存在差异;(2)LC3在AD转基因小鼠脑中的表达随Aβ表达的增加而增多,提示LC3在AD的发病中发挥作用。
Objective To observe the expressions of β-amyloid(Aβ) and Microtubule-associated protein light chain 3(LC3),the marker of autophagy,in some memory-associated brain regions of 5 × FAD mice, a transgenic model for Alzheimer's disease(AD). Moreover, we investigate the relationship between expression quantities of these two proteins. Methods Paraffin-embedded slides, immunohisto- chemistry, photomicrography and stereology were used to detect expressions of Aβ and LC3. Data analyses were performed with the SPSS software version 11.5. Result (1) Compared to wild-type mice, the common expressions of Aβ and LC3 were both increased in 5 × FAD mice(P〈0.05). (2) The expression level of Aβ and LC3 in specific brain regions of mutant type, such as M1, M2, GD, CA1, CA3, CPu were higher than in the corresponding regions of wild type. (3) No significant differences were found among the GD, CA1 and CA3 regions of hippocampus on the expression level of Aβ and LC3 in mutant mice. There were also no significant differences among the M1, M2 and hippocampus on the expression level of Aβ and LC3 in brains of mutant type. However, the expression levels of Aβ and LC3 in above regions of mutant brains were higher than those in CPu. While the expression level of Aβ and LC3 in Cb were the least. (4) The correlation between increased LC3 and deposit of Aβ was confirmed in 5XFAD mice(r= 0. 486, P〈0.05). Conclusion (1) Common expressions of Aβand LC3 are both increased in 5 ×FAD mice. Furthermore, the increases of their expressions among different memory associated brain regions are not on the same level. By contrast, they apt to deposit in CPu, and hardly deposit in Cb. (2) The expression of LC3 were upregulated with the increased of Aβ expression in 5 × FAD mice, suggested that LC3 play a role in the process of AD.
