摘要
信号传导与转录因子家族(STATs)在人体内发挥着信号传导和转录因子双重作用,STAT3在癌症、炎症和干细胞再生等过程中起着重要作用,使之成为最受关注和研究最多的成员。目前STAT3抑制剂得到快速发展,STAT3抑制剂主要针对SH2结构域、DNA结合域或者是N-末端结构域,从而抑制与STAT3信号传导相关过程,如STAT3磷酸化、STAT3二聚化、STAT3与DNA的结合和STAT3靶基因的表达。小分子抑制剂是STAT3抑制剂数量最多的一类。
Signal transducers and activators of transcription(STATs) play an important role in the signal transduction and transcription.It has important function in cancer progression,inflammation,stem cell self-renewal and so forth,so STAT3 has received particular attention.Now STAT3 inhibitors get rapid development in many areas.STAT3 inhibitors target one of the SH2 domain,DNA binding domain and N-terminal domain,which lead to the suppression of STAT3 phosphorylation,STAT3 dimerization,STAT3-DNA binding,expression of STAT3 target genes and so forth.Small-molecule inhibitors compose the largest class of STAT3 inhibitors.
