摘要
目的:考察丁香挥发油、肉豆蔻挥发油、八角茴香挥发油对磷酸川芎嗪透皮贴剂的促渗作用。方法:以小白鼠为透皮屏障,3%月桂氮卓芯酮为阳性对照,每种挥发油分别考察3%,5%,7%的体积分数对磷酸川芎嗪透皮贴剂的经皮渗透的影响,筛选出同种挥发油的不同体积分数中5%丁香油、3%八角茴香油、3%肉豆蔻油对磷酸川芎嗪透皮贴剂的经皮渗透效果最好。采用改良Franz扩散池,以裸鼠皮肤为透皮屏障,HPLC法测定磷酸川芎嗪的累积渗透量,验证以小白鼠为透皮屏障筛选出的最佳挥发油。结果:磷酸川芎嗪12 h的累积渗透量(Q12)大小依次为5%丁香油>3%月桂氮卓芯酮>阴性对照>3%八角茴香油>3%肉豆蔻油。5%丁香油促透效果最好,12 h的累积渗透量可达3.964 mg.cm-2,12 h增渗倍数为3.22,与阴性组相比差异有非常显著性意义(P<0.01),且高于阳性对照组月桂氮卓芯酮(Q12为3.056 mg.cm-2;ER12为2.23)。结论:丁香油对磷酸川芎嗪透皮贴剂的促渗作用显著,肉豆蔻油、八角茴香油未观察到促渗作用。
OBJECTIVE To investigate the effect of volatile oils,which were extracted from Flos Caryophylli(FC),Semen Myristicae(SM),Fructus Anisi Stellati(FAS),on the permeation of ligustrazine phosphate(LP) patches through hairless mouse skin in vitro.METHODS A test of penetration through the skin was conducted by using an improved method of Franz diffusion cells in preliminary experiments.The effect of volatile oil of FC,SM and FAS on the permeation degree of LP from the concentration of 3%,5%,7% by each volatile oil was compared with 3% azone.As a result the best enhancing effect by each volatile oil was 5% volatile oil of FC、3% volatile oil of FAS and 3% volatile oil of SM.A test of penetration through hairless skin mice was conducted by using an improved method of Franz diffusion cells.The effect of volatile oil of FC,SM and FAS on the permeation degree of LP through hairless skin mice was observed and compared with 3% azone.The content of LP was detected by using HPLC.RESULTS The sequence of average permeation quantity(Q12) of the three kinds of volatile oil was 5% volatile oil of FC3% azone negative control 3% volatile oil of FAS3% volatile oil of SM.5% volatile oil of FC had the highest enhancing effect,its average permeation quantity was 3.964 mg·cm-2,it was significantly higher(P〈0.01) than the negative group by independent-samples t-test.12 h Enhancing Rates(ER) of 5% volatile oil of FC was 3.22,it was better than 3% azone(Q12 was 3.056 mg·cm-2,12 h ER was 2.23).CONCLUSION The volatile oil of FC could enhance penetration of LP obviously,but the enhancing effects of volatile oil of SM and FAS were not observed.
