摘要
目的探讨壮骨镇痛胶囊对乳腺癌裸鼠移植瘤的抑制作用及机制。方法通过建立乳腺癌裸鼠移植瘤模型,将32只裸鼠随机分为壮骨镇痛胶囊治疗组、壮骨镇痛胶囊预防组、参一胶囊阳性药物对照组和模型组。观察各组裸鼠移植瘤的生长状况,测量移植瘤体积,绘制生长曲线,计算抑瘤率,免疫组化检测各组瘤体组织血管内皮生长因子(VEGF)和基质金属蛋白酶-9(MMP-9)表达。结果壮骨镇痛胶囊预防组、治疗组和参一胶囊组的移植瘤体质量明显低于模型组,差异有统计学意义(P<0.01)。壮骨镇痛胶囊治疗组与参一胶囊组瘤重比较差异无统计学意义(P>0.05)。与模型组比较,壮骨镇痛胶囊预防组、治疗组及参一胶囊组瘤体内VEGF和MMP-9的表达均明显下调(P<0.01)。与壮骨镇痛胶囊治疗组及参一胶囊组相比,壮骨镇痛胶囊预防组瘤体内VEGF和MMP-9下调更显著(P<0.01)。壮骨镇痛胶囊治疗组与参一胶囊组比较瘤体内VEGF和MMP-9表达水平无明显差异。结论壮骨镇痛胶囊可能通过下调肿瘤组织VEGF和MMP-9表达,抑制内皮细胞增殖和迁移而抑制血管生成,进而抑制高转移性乳腺癌生长。
Objective To discuss the inhibition effects and the mechanism of Zhuanggu Zhengtong capsules(ZZC) on breast carcinoma xenografts in nude mice.Methods Models of xenograft breast carcinoma in nude mice were established and 32 modeled mice were randomly divided into four groups as follows: ZZC treatment group,ZZC preventative group,ShenYi capsule(SYC) positive group and model group.Dynamic changes of transplanted tumors were observed,the volumes of the transplanted tumors were measured,the growth curves were plotted and inhibition rates were calculated.VEGF and MMP-9 expression in tumor tissues of different groups were detected by immunohistochemistry.Results Compared with model group,the weight of the xenografts of breast carcinoma in ZZC treatment group,ZZC preventative group and SYC group was reduced markedly(P〈0.01).There was no significant difference in the tumor weight between ZZC treatment group and the SYC positive group(P〈0.05).Compared with model group,the expression levels of VEGF and MMP-9 in the ZZC treatment group,the ZZC preventative group,the SYC positive group were markedly reduced.The reduction in the ZZC preventative group was more obvious than those of other medicine groups while no significant differences between the other two medicine groups.Conclusion ZZC may suppress the proliferation and migration of endothelial cells by down-regulating VEGF and MMP-9 expression in the tumor tissues,which results in the inhibition of the angiogenesis,and then inhibits the growth of highly metastatic breast carcinoma.
