摘要
对欧洲鹅耳枥种子变温层积过程中种皮和胚的动态变化分别进行扫描电镜和透射电镜观察,结果表明,随着层积时间的增长,种子外种皮破损、出现裂缝至完全脱落;中种皮出现明显孔洞、裂缝至完全疏松,内种皮细胞排列越来越疏松,透性加大,利于种子的萌发。暖层积后,种胚内脂类物质及高电子致密物质明显减少,出现降解、消融现象,表明暖层积促进贮藏物质的转化;进入冷层积后贮藏物质继续降解、消融,且出现少量淀粉粒、线粒体等,表明种子休眠逐渐解除。
We observed the dynamic changes of the seed coat and embryo of Carpinus betulus under scanning and transmission electron microscope in different period of fluctuating temperature stratification systematic.With the stratification time,the exopleura of the Carpinus betulus seed is damaged,and there are some cracks leading to the completely abscission of the exopleura finally.There are some obvious holes in mesosperm of the Carpinus betulus seed,and then the mesosperm completely looses.Endotesta cells of the Carpinus betulus seed become looser and the permeability of endotesta cells become stronger.All these factors contribute to the seed germination.After warm stratification,the lipid body and black granule in the seed embryo gradually decrease obviously,and even disappear finally.The warm stratification is important for the transformation of storage substance in the seed embryo.In the cold stratification,the Carpinus betulus seed continues degrading and some starch grain and mitochondria appear.The seed dormancy of Carpinus betulus seed is overcome gradually.
