摘要
目的探讨人脐间充质干细胞(UC-MSCs)治疗子痫前期的可行性。方法 1)应用DMEM培养人脐静脉内皮细胞(HUVEC),分4组(每组n=30)干预:分别应用含15%健康孕妇血清、含15%子痫前期轻度患者血清、含15%子痫前期重度患者血清(24 h尿蛋白>2~<4 g,简称低蛋白组)及含15%子痫前期重度患者血清(24 h尿蛋白>4 g,简称高蛋白组)在37℃、5%CO2培养箱培养24 h,处理后应用Transwell技术建立内皮细胞通透模型并计算通透系数;2)分别取上述干预后4组细胞的上清液,处理后加入Transwell小室下室,将第5代人脐间充质干细胞以1×105/mL接种到Transwell小室的上室,在37℃、5%CO2培养箱培养24 h后,于200倍镜下随机计数6个无重叠细胞视野的透膜细胞数,取其平均数代表该组细胞的侵袭能力。结果 1)4组的通透系数[Pa(%)]分别为:110.3±2.92、120.7±2.93、141.4±1.81、169.8±2.63(P<0.05)。2)4组透膜细胞数分别为(个):11.00±2.71、24.10±2.92、35.32±6.03、47.00±1.93(P<0.05)。结论子痫前期患者血清对脐静脉内皮细胞有明显损伤,损伤的内皮细胞会对产生趋化作用,而这种趋化作用随着随损伤程度的加重而增强。UC-MSCs移植有可能成为治疗子痫前期的方法之一。
Objective To study the feasibility of UC-MSCs transplantation in vitro for the treatment of preeclampgia.Methods 1 )Human umbilical vein endothelial cells (HUVEC) were planted , then incubated by DMEM' with different contents of sermn,which was divided into 4 groups( n = 30) ,that is 15% serum of healthy pregnant women, 15% serum of patients with mild preeclampsia, 15% serum of patients with severe preeclampsia (whose 24 h urine protein 〉 2 - 〈 4 g), and15% serum of patients with severe preeclampsia (whlose 24 h urine protein 〉 4 g). EndotheliM permeability was meas- ured. 2) Collected the culture solution in a sterile EP of the above 4 groups, then added the solution into the lower chamber of Transwell. The upper chamber of Transwell was filled with the 5th passage of UC-MSCs ( 1 × 105/mL) . After 24 h of in- cubation in 5% CO2 at 37℃ ,the extent of invasion was determined by counting the number of stained cens on the mem- branes in 6 randomly selected, non-overlapping fields at 200 magnification under a light microscope. Results 1 ) The per- meability(Pa% ) of 4 groups were 110.3 ±2.92,120.7 ±2.93,141.4±1.81 and 169.8 ±2. 63(P 〈0.05). 2) The num- ber of stained ceils on the membranes of 4 groups were 11.00 ±2. 71,24. 10 ±2. 92,35.32 ±6. 03 and 47. 00 ±1.93 ( P 〈 0. 05). Conclusion The serum of patients with preeclampsia could induce a significant increase in the EndotheliaI per- meability,the effect on HUVE cells.could increase the chemotactic characteristic of UC-MSCS to vascular endothelial cells incubated.
