摘要
目的探讨巨噬细胞移动抑制因子(MIF)rs1007888位点基因多态性与妊娠期空腹血糖(FBG)及胰岛素抵抗的相关性。方法选取2011年7月—2012年7月我院门诊和住院收治的孕妇240例,其中包括妊娠期糖尿病(GDM)孕妇110例,正常孕妇130例。空腹6h后晨起采集所有孕妇的外周静脉血并提取DNA,采用PCR和DNA测序技术检测MIF rs1007888位点基因型;测定FBG和空腹胰岛素(FIN),并采用稳态模型评估法计算胰岛素抵抗指数(HOMA-IR),进而比较不同基因型孕妇FBG及HOMA-IR的差异。结果 MIF rs1007888位点共检测出GG、GA、AA等3种基因型。GG基因型孕妇与GA、AA基因型孕妇的FBG及HOMA-IR比较,差异均有显著性(F=37.88、40.26,q=3.48~4.01,P<0.05)。结论 MIF rs1007888位点基因多态性与妊娠期FBG升高及胰岛素抵抗有关,GG基因型孕妇可能更易患GDM。
Objective To investigate the correlation between rs1007888a single nucleotide polymorphisms in macrophage migration inhibitory factor (MIF) gene and gestational fasting blood glucose (FBG) and insulin resistance (HOMAqR). Methods This study consisted of 240 pregnant women who were hospitalized in our hospital during 2011.11 2012.07. Of whom, 110 with gestational diabetes mellitus (GDM) and 130 healthy pregnant. MIF rs1007888 locus genotypes were detected by PCR and DNA sequencing technology in fasting blood samples. The levels of FBG and fasting insulin (FIN) were determined, and HOMA IR was quantified by homeostasis model assessment. The differences of FBG and HOMA IR were compared between different-ge- notype pregnant women. Results There were three genotypes detected at MIF rs1007888: GG, GA, and AA. Differences of FBG and HOMA IR between GG genotype and GA-and AA genotype pregnant women were significant (F = 37.88,40.26;q = 3.48--4.01 ;P〈0.05). Conclusion MIF polymorphism rs1007888 is associated with high gestational FBG and HOMA IR. GG genotype pregnant women are more liable to suffering from diabetes.
