摘要
目的通过对牡丹皮及其不同炮制程度炭品吸附力和鞣质含量的测定,探索建立牡丹皮炭质量的标准。方法利用牡丹皮及其不同炮制程度炭品对色素亚甲基蓝的吸附情况以及磷钼钨酸与其中所含的多酚类物质的显色反应,采用紫外-可见分光光度法测定相应吸光度,用标准曲线法计算吸附力及鞣质含量。结果亚甲基蓝在0.011 98~0.119 80 mg时线性关系良好;没食子酸在0.000 200 8~0.010 040 0 mg/mL时线性关系良好。牡丹皮、轻炭、标炭、重炭吸附力分别为(7.454±0.083)、(7.630±0.046)、(7.786±0.039)、(7.605±0.061)mg/g;牡丹皮、轻炭、标炭、重炭鞣质含量分别为(7.754±0.118)、(3.451±0.131)、(0.896±0.082)、(0.397±0.091)mg/g。结论建议牡丹皮炭的色素对亚甲基蓝的吸附力不低于7.7 mg/g、鞣质含量不低于0.81 mg/g。建立的牡丹皮炭质量评价标准科学合理、简便可行。
Objective To establish the quality standard of Cortex moutan carbon by detecting the adsorption capacity and the tannin content in cortex moutan as well as its different processing degrees of carbons. Methods By using the absorption of methylene blue pigment by Cortex moutan and its different processing degrees of carbon and the chromogenic reaction of tungsten molybdophosphate with the contained polyphenol materials,the corresponding absorbance degree was detected by the ultraviolet visible spectrophotometric method,the adsorption capacity and the tannin content were calculated by the standard curve method. Results The linear relationship was good when methylene blue was 0.011 98-0.119 80 mg or gllic cid was 0.000 200 8-0.010 040 0. The absorption capacity of cortex moutan,cortex moutan light carbon,cortex moutan standard carbon and cortex moutan heavy carbon was (7.454±0.083)mg/g, (7.630±0.046)mg/g, (7.786±0.039)mg/g, (7.605±0.061)mg/g respective- ly;tannin content of cortex moutan, cortex moutan light carbon, cortex moutan standard carbon and cortex moutan heavy carbon was(7.754±0.118) mg/g, (3.451±0.131)mg/g, (0.896±0.082)mg/g, (0.397±0.091)mg/g respectively. Conclusion The ab- sorption capacity of pigment to methylene blue in cortex moutan should not below 7.7 mg/g and the tannin content should not be below 0.81 mg/g. The established quality evaluation standard of cortex moutan carbon is scientific, rational, simple, convenient and feasible.
