摘要
目的:研究茅苍术醇提取物在子宫颈癌SKOV-3细胞的抗瘤效果。方法:90%乙醇超声提取茅苍术1h后,离心收集上清液;MTT比色法检测不同浓度茅苍术醇提取物对SKOV-3细胞增殖的抑制效果;流式细胞术检测茅苍术刺激SKOV-3细胞的细胞周期变化;荧光定量RT-PCR检测茅苍术刺激前后细胞周期蛋白D1(cyclin D1)mRNA表达量变化。结果:茅苍术醇提取物在5~100μg/ml之间能有效抑制SKOV-3细胞的增殖,且抑制效果与茅苍术醇提取物浓度呈现依赖性,茅苍术刺激48h后,抑制效果最佳,IC50约为70.42μg/ml;PI单染显示茅苍术刺激后,将细胞周期阻滞在G0/G1期;cyclin D1 mRNA表达因茅苍术刺激出现下调。结论:茅苍术醇提取物能有效抑制cyclin D1的表达,进而将细胞周期阻滞于G0/G1期,最终实现其抗肿瘤作用效果。
To research the anti-tumor effect of ethanol extract of Rhizema Atractylodis on SKOV-3 ceils. Methods: Rhizema Atractylo- dis was extracted by 90% ethanol using ultrasonic method for 1 h, then supematnt was collected by centrifugation; The proliferation of SHOV- 3 inhibited by ethanol extract of Rhizoma Atractylodis was measured by MTY assay. The effect of ethanol extract of Rhizoma Atractylodis on cell cyclic was detected by flow cytometry; cyclin D1 mRNA expression was measured by real-time RT-PCR. Results: Ethanol extract of Rhi- zoma Atractylodis (51,t,g/ml - 100μg/ml) can effectively inhibit the proliferation of SKOV-3 in dosage and time dependent manner, the best inhibitory effect was appeared on 48h, the IC50 is about 70.42μg,/ml; ethanol extract of Rhizema Atractylodis can block cell cyclic in G0/G1 phase, and decrease eyclin D1 mRNA expression. Conclusion: Ethanol extract of Rhizoma Atractylodis can block ceil cyclic in G0/G1 phase via decrease cyclin D1 mRNA expression to realize anti-tumor effect.
