摘要
目的观察大黄素甲醚联合人脐带间充质干细胞(HUC-MSCs)移植对颅脑损伤大鼠脑组织MMP-9和TIMP-1蛋白酶的影响。方法将40只2-3月龄,体重200~250g,清洁级Wistat大鼠随机分为正常对照组(10只)、TBI组(10只)、HUC-MSCs移植组(10只)、HUC-MSCs移植+大黄素甲醚组(10只)。ELISA法检测基质金属蛋白-9(mat rix metalloproteinase-9,MMP-9)和金属蛋白酶组织抑制物-1(tissue inhibi-tor of metall oproteinase-1,TIMP-1)的表达。结果与模型组比较,HUC-MSCs移植组、HUC-MSCs移植+大黄素甲醚组大鼠脑组织中MMP-9表达减弱,TIMP-1表达增强。结论大黄素甲醚可使脑损伤大鼠微血管基底通透性降低,减轻脑水肿程度,并与HUC-MSCs有协同作用,其作用机制可能与增加TIMP-1表达以调节MMP-9平衡有关。
Objective To observe the effect of physcion combined with human umbilical cord mesenchymal stem cell(HUC-MSCs) transplantation on the brain tissues matrix metalloproteinases-9(MMP-9) and tissue inhibitor of metalloproteinase-1(TIMP-1) protease in rats with brain injury.Methods A Total of 40 wistat rats(aging 2-3 months,weighting 200-250g) were randomly divided into normal control group(10 rats),TBI group(10 rats),HUC-MSCs group(10 rats),transplantation of HUC-MSCs+emodin group(10 rats).ELISA assay was used to detect the expression of MMP-9 and TIMP-1.Results Compared with the model group,the expression of MMP-9 was reduced and TIMP-1 expression was increased in rats' brain tissues in the HUC-MSCs transplantation group and the HUC-MSCs transplantation+physcion group.Conclusion Physcion can decrease the microvessel permeability,reduce brain edema in brain injury rats,which is also synergistically related with HUC-MSCs.This mechanism may be associated with the increased expression of TIMP-1 for the balance regulation of MMP-9.
