环孢素对大鼠葡萄糖代谢及β细胞功能的作用

摘要 给大鼠环孢素(CsA)20mg/kg,2次/d灌胃共2周,与空白对照组比较,空腹及葡萄糖(GS)灌胃后各时相血浆GS浓度及其曲线下面积(AUC)均增高,其中1 h血浆GS及其AUC明显增高(P<0.01);空腹血浆胰岛素(In)水平无明显变化,GS灌胃后各时相血浆In水平及其AUC均下降,其中1 h血浆胰岛素及其AUC与对照组比较明显下降(P<0.001和0.05);胰腺组织In含量及体外培养In释放也明显下降(P<0.01和0.001)。结果表明经口给予大鼠CsA可损害葡萄糖代谢及β细胞功能。 The effects of cyclosporine A (CsA) on glucose metabolism and betancell functions in vivo in rats were investigated. CsA (20mg/kg, 2/d) was given to Spraguer-Dawley rats for 14d. After a 1.5g/kg body weight glucose load by gavage the plasma glucose at 1 h and the area under curve (AUC) of plasma glucose were higher in the CsA-treated group than in the control (P<0.01). The plasma insulin at 1 h and the AUC of plasma insulin were lower in the CsA-treated group than in the control (P< 0.001 and 0.05, respectively). Insulin content and insulin release in pancreases in vitro were significantly lower in CsA-treated group than in the control (P<0.001 and 0.01, respectively). These data suggest that oral administration of CsA may damage glucose metabolism and islet betancell functions.
