摘要
目的建立太空莲叶中荷叶碱含量测定方法,测定太空莲叶不同生长期和采收期荷叶碱的含量。方法采用色谱柱Hypersil BDS C18(4.6 mm×250 mm,5.0μm),乙腈-水-三乙胺-乙酸(27∶70.6∶1.6∶0.78),流动相流速为1.0 mL/min,检测波长为270 nm。结果在该色谱条件下,荷叶碱分离度良好,线性范围0.01-0.2 mg/mL。不同生长期和不同采收期荷叶中荷叶碱含量差异明显,8月初的初生嫩叶荷叶碱含量最高。结论不同采收期的近成熟叶荷叶碱含量普遍大于成熟叶,8月至9月的近成熟叶荷叶碱含量高于其他时间段的近成熟叶,因此综合考虑建议最佳采集时间为8月至9月,实验结果为荷叶资源的开发利用提供了参考依据。
Objective To establish a HPLC method for determination of nuciferine contents in space lotus leaf,and determine the content of nuciferine at different growth stages and harvest time.Methods The Hypersil BDS C18 column(4.6 mm×250 mm,5.0 μm) was used with acetonitrile-water-triethylamine-galcial acetic acid(27:70.6:1.6:0.78) as mobile phase,the flow rate was 1.0 mL/min and the UV detection wavelength was 270 nm.Results Under the chromatographic conditions,nuciferine was well separated and showed a good linear relationship at the range of 0.01 -0.2 mg/mL.The contents of nuciferine were significantly different at different growth stages and different harvest time,and were highest at early august in newborn leaves.Conclusion The content of nuciferine in nearly mature leaves was more than that in mature leaves.The content of nuciferine in nearly mature leaves at august and septemper was more than that at other month,so that the nearly mature leaves at august and septemper were best for harvest.These results may be useful for the development and utilization of Lotus leaf resources.
