维药罗勒有效提取物在体外对细胞内花生四烯酸代谢途径的影响 被引量:5

The effects of polar extracts of Uyghur Ocimum basilicum L.on the Arachidonic acid metabolic pathway in vitro

摘要 目的通过体外实验,研究维药罗勒(Ocimum basilicum L.OBL)有效提取物对花生四烯酸代谢途径的影响,从而进一步说明其抗炎作用机制。方法将用OBL-乙酸乙酯和OBL-正丁醇依次对LPS处理的RAW264.7细胞进行干预,采用ELISA法测定并比较上清液中LTB4和PGE2的浓度,求出其抑制率。结果 OBL-乙酸乙酯和OBL-正丁醇提取物对LTB4和PGE2均有不同程度的抑制作用,对LTB4的最大抑制率分别为42.78%、38.63%,对PGE2的最大有抑制率分别为57.95%、51.0%;对LTB4和PGE 2的抑制作用中OBL-乙酸乙酯的强于OBL-正丁醇(P<0.05);相同浓度的药物对PGE2的抑制率大于LTB4(P<0.05)。结论维药罗勒可能是一种双重抑制药,它通过抑制环氧合酶-2和5-酯氧化酶的活性而发挥抗炎作用。 Objective In vitro experiments to study the polar extracts of Uyghur Ocimum basilicum L.on arachidonic acid metabolic pathway,thus further its anti-inflammatory mechanism.Methods The LPS-induced RAW264.7 cells were interfered with OBL-ethyl acetate and OBL-butanol,the concentration of LTB4 and PGE2 in the supernatant was measured by ELISA and calculated the inhibition rate.Results The concentration of LTB4 and PGE2 were inhibited by OBL-ethyl acetate and OBL-butanol extract in varying degrees,the maximal inhibition ratio on LTB4 were 42.78% 、38.63%,the maximal inhibition ratio on PGE2 were 57.95% 、51.0% respectively;the inhibition rates of ethyl acetate part was stronger than the OBL-butanol(P 0.05);the inhibition rates on PGE2 was higher than LTB4 in(P 0.05) at the same dose.Conclusion OBL may be a dual inhibitor,which play an anti-inflammatory effects by inhibiting the activity of the enzyme cyclooxygenase-2 and 5-ester oxidation.
