黄芪提取物对鲤急性肝组织损伤的保护作用 被引量:6

摘要 黄芪提取物(Astragalus radix extract,ARE)是一种具有保肝作用的中草药,为了探讨黄芪提取物对鲤肝组织的保护作用,本试验用含黄芪提取物(1.0 g/kg、1.5 g/kg和3.0 g/kg)的饲料饲喂鲤60 d,再腹腔注射30%四氯化碳(CCl4)混合液,以不饲喂黄芪提取物,仅腹腔注射30%CCl4为CCl4对照组,然后抽血及采集肝和脾组织,测定肝和脾指数及血清和肝组织匀浆中谷丙转氨酶(GPT)活性、谷草转氨酶(GOT)活性、乳酸脱氢酶(LDH)活性、超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)活性、过氧化氢酶(CAT)活性、总蛋白质(TP)含量、白蛋白质(Alb)含量、总抗氧化能力(T-AOC)、谷胱甘肽(GSH)含量和丙二醛(MDA)含量,并观察肝组织病理切片。研究结果显示:1.5 g/kg和3.0 g/kg黄芪提取物组鲤血清中GPT、GOT、LDH活性均显著低于CCl4对照组(P<0.05),而其鲤血清中TP和Alb含量均显著高于CCl4对照组(P<0.05);3.0 g/kg黄芪提取物组鲤肝组织匀浆中GSH含量、CAT活性、GSH-Px活性、T-AOC和SOD活性均显著高于CCl4对照组(P<0.05),而其MDA含量均显著低于CCl4对照组(P<0.05);3.0 g/kg黄芪提取物组鲤肝、脾指数显著低于CCl4对照组(P<0.05)。病理组织切片观察结果显示,CCl4对照组肝组织出现严重的脂肪变性,而黄芪提取物处理组肝组织这种损伤明显减轻。表明黄芪提取物对鲤肝脏有保护作用,可防治鱼类肝损伤。 Astragalus radix extract (ARE)is traditionally used as herbal medicine for protecting liver from various hepatotoxic agents. To evaluate the inhibitive effects of ARE on acute liver injury in common carp, carps were fed with the di- et with 0 g/kg(control), 1.0 g/kg, 1.5 g/kg and 3.0 g/kg ARE for 60 days, respectively. After CC14 injection for 72 h, blood, liver and spleen tissue samples were collected to detect liver and spleen indexes, glutamate pyruvate transaminase (GPT) activity, glutamate oxalate transaminase (GOT) activity, lactate dehydrogenase (LDH) activity, total protein (TP) and albumin (Alb) contents, superoxide dismutase (SOD) activity, glutathione peroxidase (GSH-Px) activity, catalase ( CAT ) activity, glutathione ( GSH ) content, total antioxidant capacity ( T-AOC ) and malondialdehyde (MDA) content. Compared to control group, the activities of GPT, GOT, and LDH in serum in 1.5 g,/kg and 3.0 g/kg ARE groups were significantly decreased (P〈0. 05 ), and the contents of TP and Alb in the serum were significantly increased (P〈0. 05). The activities of CAT, SOD and GSH-Px, GSH content and T-AOC in liver in 3.0 g/kg ARE group were significantly increased (P〈0. 05), and MDA content droped (P〈0. 05). The lever and spleen indexes in carp liver feeding on 3.0 g/kg ARE were significantly lower than those in control group (P〈0. 05). Severe fatty degeneration was observed in control group which was alleviated in ARE groups. In conclusion, ARE could be used as a hepatoprotective agent for fish liver injury.
