头颈部鳞癌中ALDH1、α-SMA的表达及其意义

摘要 目的研究头颈部鳞癌(HNSCC)中ALDH1和α-SMA的表达情况及其与肿瘤转移和分期的关系。方法通过免疫组化双染在60例HNSCC标本上对ALDH1和α-SMA的表达水平和分布情况进行研究,同时利用Real-time PCR对其mRNA表达情况进行半定量分析比较,结合病例临床病理资料研究加以分析。结果 ALDH1在HNSCC中有不同程度的表达,其表达水平与头颈肿瘤分期及淋巴结转移间关系紧密,并具有统计学意义;α-SMA阳性的细胞主要分布与肿瘤-正常组织交界处,其表达强弱与淋巴结转移与否密切相关,α-SMA在分化差的病例中表达有增高趋势但无统计学意义;ALDH1和α-SMA的表达在有转移的肿瘤中具有高度协同性。结论 ALDH1和α-SMA可分别或相互组合作为判断HNSCC有无转移的指标。 Objective To investigate the expression of ALDH1 andα-SMA in head and neck squamous cel carcinoma (HNSCC) and its clinicopathological significance. Methods The clinical and pathological data of 60 cases of HNSCC were col-lected. The expression of ALDH1 andα-SMA proteins was examined by immunohistochemical double staining on paraffin sec-tions;and the ALDH1 andα-SMA mRNA levels were determined by semi-quantitative real-time PCR. Results The expression of ALDH1 was closely correlated with tumor stage and lymph node metastasis in HNSCC. Theα-SMA positive cel s were mainly located at the invading border of tumor and its expression level was closely correlated with lymph node metastasis. There was a tendency of increasedα-SMA positive cel s in poorly-differentiated cancers;however, there was no statistical difference. ALDH1 andα-SMA expression showed a high coincidence in tumors with positive lymph nodes. Conclusions ALDH1 and/orα-SMA might be good indicators for prediction of lymph node metastasis in HNSCC.
